胰腺癌中Ring1B、LSD1及P16表達(dá)及其與預(yù)后的關(guān)系

【摘要】:目的:探討胰腺癌中E3泛素連接酶亞單位Ring1B、組蛋白賴氨酸特異性去甲基化酶1(LSD1)及細(xì)胞周期調(diào)節(jié)因子P16表達(dá)與及其與患者預(yù)后的關(guān)系。方法:用免疫組化法檢測85例胰腺癌患者手術(shù)標(biāo)本(癌組織與癌旁組織)中LSD1、Ring1B與P16蛋白的表達(dá)及分布,并選取其中6對癌組織和癌旁組織,用qRT-PCR和Western blot法檢測mRNA和蛋白表達(dá);分析三者在胰腺癌組織中表達(dá)的相關(guān)性,及其與患者生存的關(guān)系。結(jié)果:免疫組化結(jié)果顯示,Ring1B與LSD1蛋白主要表達(dá)于細(xì)胞漿,P16主要表達(dá)于胞核;Ring1B與LSD1在胰腺癌組織中表達(dá)明顯高于癌旁組織,而P16的表達(dá)明顯低于癌旁組織(均P0.05);在胰腺癌組織中Ring1B、LSD1的表達(dá)與P16的表達(dá)均呈負(fù)相關(guān)(r=-476;r=-0.673,均P0.05)。qRTPCR結(jié)果顯示,胰腺癌組織中Ring1B和LSD1的mRNA的平均相對表達(dá)量均明顯高于癌旁組織(8.908vs.1.947;7.126 vs.1.940,均P0.05),P16的mRNA的平均相對表達(dá)量明顯低于癌旁組織(1.269vs.5.237,P0.05);Western blot結(jié)果顯示,三者的蛋白表達(dá)趨勢與mRNA表達(dá)一致。生存分析顯示,Ring1B、LSD1高表達(dá)患者生存率均分別低于其低表達(dá)患者(χ~2=8.958,P=0.012;χ~2=8.856,P=0.010),而P16高表達(dá)患者生存率高于其低表達(dá)患者(χ~2=7.867,P=0.024)。結(jié)論:胰腺癌組織中Ring1B與LSD1表達(dá)升高,P16表達(dá)降低;Ring1B、LSD1可能分別通過組蛋白泛素化和去甲基化調(diào)控P16的表達(dá),從而影響胰腺癌預(yù)后。
[Abstract]:Aim: to investigate the relationship between the expression of E 3 ubiquitin ligase subunit Ring1B, histone specific demethylase 1 (LSD1) and cell cycle regulator P16 in pancreatic cancer and their relationship with prognosis. Methods: immunohistochemical method was used to detect the expression and distribution of LSD1,Ring1B and P16 protein in 85 patients with pancreatic cancer. The expression of mRNA and protein was detected by qRT-PCR and Western blot. To analyze the relationship between the expression and survival of pancreatic cancer. Results: the results of immunohistochemistry showed that Ring1B and LSD1 were mainly expressed in cytoplasm and P16 was mainly expressed in nucleus, Ring1B and LSD1 in pancreatic carcinoma were significantly higher than those in paracancerous tissue, and the expression of P16 was significantly lower than that in paracancerous tissue (P0.05). There was a negative correlation between the expression of Ring1B,LSD1 and the expression of P16 in pancreatic carcinoma (r-476). The average relative expression of Ring1B and LSD1 in pancreatic cancer tissues was significantly higher than that in adjacent tissues (8.908vs.1.947). The average relative expression of mRNA in P16 was significantly lower than that in paracancerous tissues (1.269 vs.5.237); Western blot). The results showed that the protein expression trend of P16 was consistent with that of mRNA. Survival analysis showed that the survival rate of patients with high expression of Ring1B,LSD1 was lower than that of patients with low expression (蠂 ~ 2 = 8.958; P ~ + 0.010), while the survival rate of patients with high expression of P16 was higher than that of patients with low expression of P16 (蠂 ~ (2) = 7.867% P ~ (0.024). Conclusion: the expression of Ring1B and LSD1 increased and the expression of P16 decreased in pancreatic carcinoma, and Ring1B,LSD1 may affect the prognosis of pancreatic carcinoma by regulating the expression of P16 by histone ubiquification and demethylation, respectively.
