【摘要】：目的:探討DIAPH3蛋白對(duì)三陰性乳腺癌細(xì)胞遷移侵襲能力的影響;方法:本研究通過(guò)q PCR和western blot探討DIAPH3蛋白在MCF-10A、SUM159、MCF-7和MDA-MB-231細(xì)胞中的表達(dá),然后通過(guò)構(gòu)建過(guò)表達(dá)載體過(guò)表達(dá)DIAPH3,通過(guò)Transwell分析過(guò)表達(dá)DIAPH3蛋白對(duì)三陰性乳腺癌細(xì)胞遷移和侵襲能力的影響,最后探討DIAPH3蛋白影響三陰性乳腺癌細(xì)胞遷移和侵襲能力的分子機(jī)制。結(jié)果:MCF-10A、SUM159、MCF-7和MDA-MB-231中的相對(duì)表達(dá)量依次為1.00±0.000、0.47±0.068、0.64±0.039和0.17±0.008。和正常乳腺上皮細(xì)胞MCF-10A相比,乳腺癌細(xì)胞SUM159、MCF-7和MDA-MB-231中DIAPH3蛋白的相對(duì)表達(dá)量顯著降低,差異具有統(tǒng)計(jì)學(xué)意義(P0.05)。和MCF-7相比,SUM159和MDA-MB-231中DIAPH3蛋白的相對(duì)表達(dá)量顯著降低,差異具有統(tǒng)計(jì)學(xué)意義(P0.05)。Cell group、NC group和DIAPH3 group中MDA-MB-231細(xì)胞的遷移數(shù)目分別為305.5±6.36、329±16.97和221±24.04;Cell group、NC group和DIAPH3 group中MDA-MB-231細(xì)胞的侵襲數(shù)目為96±8.25、103±18.59和24.5±5.68;和Cell group、NC group相比,過(guò)表達(dá)DIAPH3能夠顯著抑制MDA-MB-231細(xì)胞的遷移和侵襲;進(jìn)一步研究表明過(guò)表達(dá)DIAPH3蛋白能夠抑制Rho A、MMP-2和MMP-9蛋白的表達(dá)。結(jié)論:DIAPH-3在乳腺癌細(xì)胞中表達(dá)顯著降低,在三陰性乳腺癌細(xì)胞中表達(dá)進(jìn)一步降低;過(guò)表達(dá)DIAPH-3能夠抑制Rho A、MMP-2和MMP-9蛋白的表達(dá),從而抑制TNBC細(xì)胞的遷移和侵襲。
[Abstract]:Objective: to investigate the effect of DIAPH3 protein on the migration and invasion of triple-negative breast cancer cells. Methods: to investigate the expression of DIAPH3 protein in MCF-10A,SUM159,MCF-7 and MDA-MB-231 cells by Q PCR and western blot. Then the overexpression vector DIAPH3, was constructed to analyze the effect of overexpression of DIAPH3 protein on the migration and invasion of triple-negative breast cancer cells by Transwell. Finally, the molecular mechanism of DIAPH3 protein affecting the migration and invasion of triple-negative breast cancer cells was discussed. Results the relative expression levels in MCF-7 and MDA-MB-231 were 1.00 鹵0.000,0.47 鹵0.068,0.64 鹵0.039 and 0.17 鹵0.008, respectively. Compared with normal breast epithelial cell MCF-10A, the relative expression of DIAPH3 protein in SUM159,MCF-7 and MDA-MB-231 of breast cancer cells was significantly lower than that of normal breast epithelial cells (P0.05). The relative expression of DIAPH3 protein in SUM159 and MDA-MB-231 was significantly lower than that in MCF-7 (P0.05). The number of migration of MDA-MB-231 cells in group,NC group and DIAPH3 group was 305.5 鹵6.36329 鹵16.97 and 221 鹵24.04Cell group,NC group, respectively, and the number of invasion of MDA-MB-231 cells in DIAPH3 group was 96 鹵8.25103 鹵18.59 and 24.5 鹵5.68, respectively, compared with Cell group,NC group. Overexpression of DIAPH3 could significantly inhibit the migration and invasion of MDA-MB-231 cells, and further studies showed that overexpression of DIAPH3 protein could inhibit the expression of Rho Agna MMP-2 and MMP-9 proteins. Conclusion the expression of DIAPH-3 in breast cancer cells was significantly lower than that in triple-negative breast cancer cells, and the overexpression of DIAPH-3 could inhibit the expression of MMP-2 and MMP-9 proteins in TNBC cells, thus inhibiting the migration and invasion of TNBC cells.
