文章摘要：腫瘤干細胞學說認為大部分腫瘤來源于腫瘤干細胞。腫瘤干細胞與正常干細胞一樣具有自我更新能力。能夠產(chǎn)生腫瘤組織團塊中的各種增殖和分化的細胞。腫瘤干細胞可能來源于正常干細胞或分化細胞。干細胞niche是干細胞生存的微環(huán)境．通過提供抑制細胞增殖和生長的信號來維持干細胞于靜止狀態(tài)。干細胞niche功能異常會導致niche的數(shù)量增加．以及干細胞功能異常和數(shù)量增加。腫瘤干細胞可能在異常的niche中存活，打破這種異常的niche就會削弱腫瘤干細胞的自我更新．從而抑制腫瘤的生長。靶向腫瘤干細胞異常微環(huán)境的治療策略可能是癌癥治療的一個方向。

Abstr：The tumor stem cell model proposes that tumor stem cells are the origin of most tumors. Like normal stem cells, tumor stem cells have the ability to self-renew and to give rise to the variety of proliferating and differentiated cells that make up the bulk of a tumor, Tumor stem cells could be derived from normal stem cells or differentiated cells. The stem cell niches are the microenvironment that maintains stem ceils primarily in a quiescent state by providing signals that inhibit cell proliferation and growth. Loss of niches can result in loss of stem cells and increasing niches lead to an increased number of stem cells. Tumor stem cells could live in abnormal niches. Disrupting this abnormal niches may impair tumor stem cell self- renewal, thereby, significantly inhibits the growth of tumors. Targeting the unique microenvironment of tumor stem cells may be the key of effective cancer therapy.
