苯丁酸鈉對多烯紫杉醇耐藥前列腺癌細胞株的增值抑制和凋亡誘導(dǎo)作用

【摘要】:目的研究苯丁酸鈉對前列腺癌多烯紫杉醇耐藥細胞株的增值抑制與凋亡誘導(dǎo)作用,并探討其作用機制。方法構(gòu)建多烯紫杉醇耐藥前列腺癌PC3細胞株(PC3/DTX),采用0、1、2、4 mmol/L的苯丁酸鈉處理PC3/DTX,MTT檢測細胞增殖、細胞活力以及抑制率,計算不同濃度苯丁酸鈉處理后各組多烯紫杉醇對PC3/DTX細胞的半致死濃度(IC50);流式細胞儀檢測不同處理組細胞凋亡水平;Western blot檢測p21、Cyclin D1與Survivin的表達水平。結(jié)果 0、1、2、4 mmol/L的苯丁酸鈉處理PC3/DTX細胞48 h后,細胞存活率分別是(99.85±2.69)%,(84.68±3.87)%,(68.65±4.54)%和(43.54±5.69)%(P0.05);抑制率分別是(10.69±3.65)%,(5.78±4.58)2%,(54.68±3.98)%和(69.84±6.54)%(P0.05);多烯紫杉醇對各處理組細胞的IC50分別為135.98±2.69、109.65±3.87、87.65±3.84、64.62±2.98 nmol/L(P0.05);細胞的凋亡率分別為(7.20±0.80)%,(10.20±0.90)%,(19.80±2.10)%和(27.40±2.50)%(P0.05);Western blot顯示苯丁酸鈉促進p21蛋白的表達,抑制Cyclin D1與Survivin蛋白的表達。結(jié)論苯丁酸鈉可以增加前列腺癌耐藥細胞株對多烯紫杉醇的藥敏性,并影響前列腺癌耐藥細胞株p21、Cyclin D1與Survivin蛋白的表達量。
[Abstract]:Aim to study the effect of sodium phenylbutyrate on proliferation inhibition and apoptosis induction of docetaxel resistant prostate cancer cell line. Methods PC3 / DTXT cell line was constructed and treated with sodium phenylbutyrate of 4 mmol / L to detect cell proliferation, cell viability and inhibition rate. The half-lethal concentration of docetaxel on PC3 / DTX cells was calculated after different concentrations of sodium phenylbutyrate, the apoptosis level of PC3 / DTX cells was detected by flow cytometry and the expression levels of p21 Cyclin D1 and survivin were detected by flow cytometry.
