PTEN在口腔白斑及口腔鱗狀細(xì)胞癌中的表達(dá)及其臨床意義

【摘要】:目的:研究第10號染色體缺失的磷酸酶及張力蛋白同源基因蛋白(PTEN)在口腔白斑和口腔鱗狀細(xì)胞癌中的表達(dá)及其臨床意義,探究其在口腔癌變中可能的機(jī)制作用。方法:收集20例口腔鱗狀細(xì)胞癌組織、20例距癌組織2cm的癌旁正常組織及20例口腔白斑組織,Western blotting測定上述3種組織中PTEN蛋白含量,qRT-PCR測定PTEN mRNA的表達(dá)水平,免疫組織化學(xué)測定PTEN蛋白在上述組織細(xì)胞中的組織學(xué)定位與表達(dá)情況。結(jié)果:1、免疫組化結(jié)果顯示PTEN蛋白多位于細(xì)胞漿中呈黃色、棕色染色。Western blotting、實時熒光定量PCR、免疫組織化學(xué)檢測發(fā)現(xiàn)PTEN蛋白和mRNA在口腔鱗狀細(xì)胞癌、口腔白斑、正常粘膜組織中的表達(dá)逐漸增高(正常粘膜組織口腔白斑口腔鱗狀細(xì)胞癌),差異具有統(tǒng)計學(xué)意義(P0.05),三種試驗方法結(jié)果較為一致。2、無異常增生白斑PTEN蛋白表達(dá)明顯高于輕度異常增生或中度異常增生,差異具有統(tǒng)計學(xué)意義(P0.05)。3、PTEN蛋白在無淋巴結(jié)轉(zhuǎn)移口腔鱗狀細(xì)胞癌中的表達(dá)明顯高于有淋巴結(jié)轉(zhuǎn)移口腔鱗狀細(xì)胞癌(P0.05)。4、PTEN蛋白在高分化鱗癌中的表達(dá)明顯較中低分化鱗癌高(P0.05)。5、不同年齡、不同性別、不同發(fā)生部位口腔鱗狀細(xì)胞癌的PTEN蛋白表達(dá)無明顯差異(P0.05)。結(jié)論:1、PTEN在口腔白斑、口腔鱗狀細(xì)胞癌和口腔正常組織中存在不同程度的表達(dá),表明PTEN可能是口腔鱗癌發(fā)生、發(fā)展、侵襲轉(zhuǎn)移過程中的一個重要因素。2、PTEN可能作為口腔癌變早期新的生物標(biāo)記物,用于口腔鱗癌的診斷、預(yù)后判斷和癌前病變惡性轉(zhuǎn)化的風(fēng)險評估,PTEN在臨床治療過程中可能會成為口腔鱗狀細(xì)胞癌的治療新靶點。
[Abstract]:Aim: to investigate the expression and clinical significance of PTEN in oral leukoplakia and oral squamous cell carcinoma, and to explore the possible mechanism of PTEN in oral carcinogenesis. Methods: 20 cases of oral squamous cell carcinoma (OSCC) and 20 cases of normal tissues adjacent to 2cm and 20 cases of oral leukoplakia were collected to detect the expression of PTEN protein and PTEN mRNA by qRT-PCR in 20 cases of oral squamous cell carcinoma tissues and 20 cases of oral leukoplakia tissues. The histological localization and expression of PTEN protein were detected by immunohistochemistry. Results the results of immunohistochemistry showed that PTEN protein was mostly yellow in cytoplasm, brown staining, Western blotting, real-time fluorescence quantitative PCR, PTEN protein and mRNA were found in oral squamous cell carcinoma and oral leukoplakia by immunohistochemistry. The expression of PTEN protein in normal mucosa tissues was increased gradually (the difference was statistically significant (P 0.05). The results of the three test methods were consistent. 2. 2. There was no abnormal proliferation of leukoplakia PTEN protein expression. Higher than mild dysplasia or moderate dysplasia, The expression of P0.05PTEN protein was significantly higher in oral squamous cell carcinoma without lymph node metastasis than that in well-differentiated squamous cell carcinoma with lymph node metastasis. The expression of PTEN protein in well-differentiated squamous cell carcinoma was significantly higher than that in well-differentiated squamous cell carcinoma. There was no significant difference in the expression of PTEN protein in oral squamous cell carcinoma (OSCC) with different sex and location (P 0.05). Conclusion the expression of PTEN in oral leukoplakia, oral squamous cell carcinoma and oral normal tissue is different, suggesting that PTEN may be the occurrence and development of oral squamous cell carcinoma. PTEN, an important factor in the process of invasion and metastasis, may be used as a new biomarker for the diagnosis of oral squamous cell carcinoma. Prognostic evaluation and risk assessment of malignant transformation of precancerous lesions PTEN may be a new target for the treatment of oral squamous cell carcinoma (OSCC).
