4種人腎細胞癌原位移植裸鼠模型建立方法的比較

【摘要】:背景與目的:腎細胞癌是最常見的腎臟惡性腫瘤,起病隱匿,惡性程度高。研究旨在建立成功率高、穩(wěn)定的人腎細胞癌原位動物造模方法。方法:采用人腎細胞癌細胞(786-0、ACHN)進行細胞懸液原位注射法、皮下成瘤后引瘤原代細胞懸液原位注射法、皮下成瘤后引瘤組織塊原位移植腎包膜下法及皮下成瘤后引瘤組織塊原位移植腎周筋膜內(nèi)法建立原位移植裸鼠模型,采用PET/CT、H-E染色、免疫組織化學(xué)染色及血清生化檢測等手段觀察成瘤率及腫瘤生長情況,評估4種方法的建模效果。結(jié)果:人腎細胞癌皮下移植瘤裸鼠模型,ACHN組成瘤率(90%)高于786-0組(30%);人腎細胞癌原位移植裸鼠模型,786-0原位注射組、ACHN原位注射組、ACHN皮下移植瘤引瘤后原代細胞原位注射組、ACHN組織塊腎包膜下包埋組和ACHN組織塊腎周筋膜內(nèi)包埋組的成瘤率分別為33%、80%、90%、100%和20%,其中以ACHN皮下成瘤后引瘤組織塊原位移植腎包膜下法成瘤率最高,影像學(xué)及病理組織學(xué)檢查結(jié)果顯示符合低分化腎細胞癌。結(jié)論:成功建立4種人腎細胞癌原位移植裸鼠模型,其中以ACHN皮下成瘤后引瘤組織塊原位移植腎包膜下法成瘤效果最佳,為腎細胞癌發(fā)病機制及靶向治療的進一步研究提供理想的動物模型。
[Abstract]:Background & objective: renal cell carcinoma (RCC) is the most common renal malignancy with hidden onset and high malignancy.The aim of this study was to establish an in situ animal model of human renal cell carcinoma (RCC) with high success rate.Methods: in situ injection of human renal cell carcinoma cell line 786-0 ACHN was performed, and the primary cell suspension after subcutaneous tumorigenesis was injected in situ.Nude mice models of orthotopic transplantation were established by orthotopic transplantation of subcutaneous tumor tissue mass under renal capsule and subcutaneous tumor tissue mass orthotopic transplantation of perirenal fascia, and PET / CTH-E staining was used.The tumor formation rate and tumor growth were observed by immunohistochemical staining and serum biochemical detection, and the modeling effects of the four methods were evaluated.Results: the tumor rate of ACHN in nude mice with subcutaneous transplanted tumor of human renal cell carcinoma was 90% higher than that in group 786-0, and that in nude mice model of orthotopic transplantation of human renal cell carcinoma was 786-0.The tumorigenic rate of the subcapsular embedding group and the perirenal fascial embedding group of ACHN tissue mass were 338090% and 20%, respectively. Among them, the rate of tumor formation was the highest in the subcapsular transplantation with ACHN subcutaneous tumorigenesis and orthotopic transplantation of the tumor tissue mass, and the rate of tumor formation was the highest in the ACHN tissue mass perirenal intrafascial embedding group, and the tumor forming rate was the highest in the subcapsular implantation group after ACHN subcutaneous tumor formation.The results of imaging and histopathology showed that it was consistent with poorly differentiated renal cell carcinoma.Conclusion: four orthotopic transplantation models of human renal cell carcinoma (RCC) were successfully established in nude mice, in which ACHN subcutaneous tumorigenesis was the most effective method for renal transplantation.To provide an ideal animal model for the further study of the pathogenesis and targeted therapy of renal cell carcinoma.
