【摘要】：已有研究表明腫瘤的發(fā)生發(fā)展與氧化還原平衡密切相關(guān),近期的研究發(fā)現(xiàn)抗氧化劑對(duì)腫瘤免疫具有復(fù)雜的調(diào)控作用。硫氧還蛋白(Trx)是一種分布廣泛的抗氧化酶,能夠還原蛋白質(zhì)的半胱氨酸殘基,使得巰基和二硫鍵之間相互轉(zhuǎn)換。調(diào)節(jié)性T細(xì)胞(Treg),也稱抑制性T細(xì)胞(Suppressor T cell),在體外和體內(nèi)都顯示出很強(qiáng)的免疫抑制活性。雖然Treg與惡性腫瘤免疫抑制的關(guān)系還在研究之中,但小鼠體內(nèi)抑制Treg的研究結(jié)果表明機(jī)體抗腫瘤免疫能力得到了顯著提升。本項(xiàng)目重點(diǎn)研究硫氧還蛋白在黑色素瘤中對(duì)Treg的調(diào)節(jié)作用及對(duì)腫瘤免疫應(yīng)答的影響。主要內(nèi)容分為三個(gè)部分:第一部分收集黑色素瘤腫瘤樣品,通過免疫組化方法確定Trx在腫瘤樣品中的表達(dá)變化;通過流式細(xì)胞分選技術(shù),檢測(cè)Treg細(xì)胞在黑色素瘤組織中的浸潤情況;第二部分通過構(gòu)建Trx過表達(dá)和敲低的黑色素瘤細(xì)胞系,在荷瘤小鼠體內(nèi)研究Trx對(duì)Treg細(xì)胞浸潤的影響;第三部分使用重組的Trx蛋白和Trx中和抗體,研究Trx促進(jìn)Treg細(xì)胞浸潤的機(jī)制。實(shí)驗(yàn)結(jié)果如下:1.Trx在人類黑色素瘤組織中高表達(dá),并與腫瘤的分級(jí)分期呈正相關(guān)性;2.體外培養(yǎng)的黑素瘤細(xì)胞可以分泌大量的Trx蛋白;3.Trx可以促進(jìn)Treg細(xì)胞在黑色素瘤中的浸潤,從而提高腫瘤的免疫抑制能力;4.在體外細(xì)胞和體內(nèi)荷瘤鼠實(shí)驗(yàn)中,Trx可以提高Treg的遷移能力;5.在體外細(xì)胞和體內(nèi)荷瘤鼠實(shí)驗(yàn)中,Trx可以促進(jìn)效應(yīng)性T細(xì)胞向Treg細(xì)胞轉(zhuǎn)化;6.在體外細(xì)胞和體內(nèi)荷瘤鼠實(shí)驗(yàn)中,Trx可以提高Treg細(xì)胞的抗凋亡能力;7.Trx抗體可以抑制Treg細(xì)胞在腫瘤局部的浸潤,增強(qiáng)抗腫瘤免疫應(yīng)答;8.Trx抗體可以降低黑色素瘤的生長速率和轉(zhuǎn)移速率。綜上所述,本研究明確了Trx在黑色素瘤中對(duì)Treg浸潤的調(diào)節(jié)作用,初步分析了Trx促進(jìn)Treg細(xì)胞浸潤的機(jī)理,明確了Trx抗體在黑色素瘤免疫應(yīng)答過程中潛在應(yīng)用價(jià)值,為黑色素瘤的治療提供了新的潛在藥物靶點(diǎn)。
[Abstract]:Studies have shown that the occurrence and development of tumor and oxidation reduction equilibrium is closely related to the recent study found that antioxidants play a role in the regulation of tumor immune complex. Thioredoxin (Trx) is a kind of widely distribution of antioxidant enzymes, which reduced cysteine residues of proteins, the conversion between thiol and regulatory two disulfide bonds. T cells (Treg), also known as suppressor T cells (Suppressor T cell), both in vitro and in vivo showed strong immunosuppressive activity. Although the relationship between Treg and malignant tumor is still on immunosuppression, but research results show that the inhibition of Treg in vivo anti-tumor immune ability has been significantly improved. The project focuses on the thioredoxin regulatory role of Treg in melanoma and effect on tumor immune response. The main content is divided into three parts: the first part of the collection of melanoma tumor Tumor samples, determine the expression of Trx in tumor samples by immunohistochemical method; through flow cytometry, the infiltration of Treg cells were detected in melanoma tissues; the second part through the construction of Trx overexpression and knockdown melanoma cell lines, the effect of Trx in tumor bearing mice infiltration the third part of Treg cells; the use of recombinant Trx protein and Trx neutralizing antibody, study mechanism of Trx promoting Treg cell infiltration. The experimental results are as follows: the high expression of 1.Trx in human melanoma tissues, and grading and staging of tumor Cheng Zhengxiang; 2. cultured melanoma cells can secrete Trx protein; 3.Trx can promote the invasion of Treg cells in melanoma, thereby enhancing tumor immune suppression ability; 4. in vitro and in vivo tumor bearing rats, Trx can enhance the migration ability of Treg in vitro; 5. fine Cellular and in vivo tumor bearing rats, Trx can be transformed to Treg cells promote effector T cells in vitro and in vivo; 6. tumor bearing rats, Trx can improve the anti apoptosis ability of Treg cells; 7.Trx antibody can inhibit Treg cells in tumor infiltration, enhanced anti-tumor immune response; 8.Trx antibody to reduce the growth rate and the transfer rate of melanoma. In summary, this study defines Trx in melanoma on the regulation of Treg invasion, preliminary analysis of the mechanism of Trx promoting Treg cell infiltration, the potential application value of Trx antibody in melanoma immune response process, provide potential drug targets the new treatment for melanoma.
