瘦素對小鼠乳腺癌調(diào)節(jié)性T細(xì)胞的影響

【摘要】:目的探討瘦素對小鼠乳腺癌調(diào)節(jié)性T細(xì)胞的影響及生物學(xué)意義。方法培養(yǎng)4T1乳腺癌細(xì)胞株并接種于Balb/c小鼠建立乳腺癌模型,將成瘤小鼠隨機(jī)分為對照組和實驗組,分別用等量生理鹽水(normal saline,NS)和瘦素(leptin)處理,監(jiān)測小鼠腫瘤的生長;流式細(xì)胞技術(shù)檢測各組Balb/c荷瘤小鼠外周血、淋巴結(jié)、脾臟中調(diào)節(jié)性T淋巴細(xì)胞的數(shù)量及比例變化;ELISA檢測各組荷瘤小鼠血清TGF-β1、IL-10含量。結(jié)果與NS組比較,實驗組腫瘤生長速度隨瘦素質(zhì)量濃度的增加而加快;外周血和脾臟中調(diào)節(jié)性T細(xì)胞比例隨瘦素質(zhì)量濃度的增加而升高(P0.05),淋巴結(jié)中調(diào)節(jié)性T細(xì)胞比例變化不明顯(P0.05),血清中TGF-β1、IL-10的含量隨瘦素質(zhì)量濃度的增加而升高(P0.05)。結(jié)論瘦素通過上調(diào)乳腺腫瘤微環(huán)境中調(diào)節(jié)性T細(xì)胞的數(shù)量比例,分泌抑制因子,抑制機(jī)體腫瘤免疫,進(jìn)而促進(jìn)小鼠乳腺腫瘤的生長。
[Abstract]:Objective to investigate the effect of leptin on regulatory T cells of mouse breast cancer and its biological significance.Methods 4T1 breast cancer cell lines were cultured and inoculated into Balb/c mice to establish breast cancer model. The tumor-bearing mice were randomly divided into two groups: control group and experimental group. The mice were treated with the same amount of normal saline and leptin to monitor the growth of tumor.The quantity and proportion of regulatory T lymphocytes in peripheral blood, lymph nodes and spleen of Balb/c bearing mice were detected by flow cytometry.Results compared with NS group, the tumor growth rate of the experimental group was accelerated with the increase of leptin concentration.The proportion of regulatory T cells in peripheral blood and spleen increased with the increase of leptin concentration, while the proportion of regulatory T cells in lymph nodes did not change significantly (P 0.05). The content of TGF- 尾 1 and IL-10 in serum increased with the increase of leptin concentration.Conclusion leptin inhibits tumor immunity by upregulating the proportion of regulatory T cells in breast tumor microenvironment, secreting inhibitory factor, and then promoting the growth of breast tumor in mice.
