【摘要】:目的:研究REGγ基因促進(jìn)人乳腺癌細(xì)胞株MDA-MB-231發(fā)生上皮-間充質(zhì)轉(zhuǎn)化的生物學(xué)作用。方法:免疫組化檢測(cè)乳腺癌組織與癌旁組織REGγ的表達(dá)差異,以及檢測(cè)轉(zhuǎn)移淋巴結(jié)中REGγ的表達(dá)水平。Western Blot和RT-PCR檢測(cè)不同人乳腺癌細(xì)胞系之間REGγ的表達(dá)差異。構(gòu)建REGγ基因過(guò)表達(dá)和干擾載體并將其導(dǎo)入MDA-MB-231細(xì)胞中,然后用RT-PCR和細(xì)胞免疫熒光分別從轉(zhuǎn)錄水平和蛋白水平上檢測(cè)上皮-間充質(zhì)轉(zhuǎn)化細(xì)胞標(biāo)記物的表達(dá)水平的變化。通過(guò)劃痕實(shí)驗(yàn)和Transwell闡釋REGγ對(duì)細(xì)胞侵襲遷移能力的影響。結(jié)果:免疫組化結(jié)果提示REGγ在乳腺癌組織和轉(zhuǎn)移淋巴結(jié)中顯著高表達(dá)。Western Blot和RT-PCR提示REGγ在MDA-MB-231細(xì)胞中表達(dá)水平較高。MDA-MB-231細(xì)胞過(guò)表達(dá)REGγ后E-鈣粘素(E-cadherin)下調(diào),而波形蛋白(Vimentin)上調(diào)(P0.01);干擾REGγ后會(huì)逆轉(zhuǎn)上皮-間充質(zhì)轉(zhuǎn)化,使細(xì)胞侵襲遷移能力減弱。結(jié)論:REGγ促進(jìn)人乳腺癌MDA-MB-231細(xì)胞發(fā)生上皮-間充質(zhì)轉(zhuǎn)化,從而促使腫瘤細(xì)胞發(fā)生侵襲轉(zhuǎn)移。
[Abstract]:Objective: to study the biological effect of REG 緯 gene on epithelial-mesenchymal transformation in human breast cancer cell line MDA-MB-231. The expression level of REG 緯 in metastatic lymph nodes was detected. Western Blot and RT-PCR were used to detect the difference of REG 緯 expression between different human breast cancer cell lines. The overexpression and interference vector of REG 緯 gene was constructed and introduced into MDA-MB-231 cells. Then the expression level of epithelium-mesenchymal transformation cell marker was detected by RT-PCR and cellular immunofluorescence at the transcriptional level and protein level respectively. The effect of REG 緯 on the ability of cell invasion and migration was explained by scratch test and Transwell. Results: immunohistochemical results showed that REG 緯 was significantly overexpressed in breast cancer tissues and metastatic lymph nodes. Western Blot and RT-PCR suggested that REG 緯 expression was higher in MDA-MB-231 cells. Ecadherin E-cadherin was down-regulated in MDA-MB-231 cells after overexpression of REG 緯. Vimentin upregulated P0.01C, and interfered with REG 緯, which reversed the epithelial-mesenchymal transformation and weakened the ability of invasion and migration. Conclusion: Vimentin 緯 can promote the epithelial-mesenchymal transformation of human breast cancer MDA-MB-231 cells and promote the invasion and metastasis of tumor cells.
