長(zhǎng)鏈非編碼RNAFENDRR對(duì)宣威肺癌XWLC-05細(xì)胞增殖、遷移及侵襲的影響

【摘要】:目的:檢測(cè)非編碼RNA FENDRR長(zhǎng)鏈在宣威肺腺癌XWLC-05株細(xì)胞與非小細(xì)胞肺癌(NSCLC)A549株細(xì)胞中的表達(dá),及其對(duì)細(xì)胞增殖、遷移及侵襲的影響。方法:實(shí)時(shí)熒光定量PCR檢測(cè)FENDRR基因在XWLC-05與A549細(xì)胞中的表達(dá);構(gòu)建過(guò)表達(dá)p EX-3-FENDRR質(zhì)粒并轉(zhuǎn)染XWLC-05細(xì)胞,慢病毒干擾LV3-FENDRR載體轉(zhuǎn)染A549細(xì)胞,實(shí)時(shí)熒光定量PCR檢測(cè)轉(zhuǎn)染后XWLC-05和A549細(xì)胞內(nèi)FENDRR的表達(dá);MTS法檢測(cè)細(xì)胞增殖;Transwell小室法檢測(cè)細(xì)胞遷移及侵襲能力的變化。結(jié)果:XWLC-05細(xì)胞中FENDRR基因表達(dá)明顯較A549細(xì)胞低,成功提高轉(zhuǎn)染p EX-3-FENDRR的XWLC-05細(xì)胞和降低轉(zhuǎn)染LV3-FENDRR質(zhì)粒的A549細(xì)胞中FENDRR m RNA水平。與對(duì)照組相比,過(guò)表達(dá)FENDRR可明顯抑制XWLC-05細(xì)胞增殖[(1.23±0.13)vs(1.46±0.25),P0.05]并降低其遷移、侵襲能力(P0.05);干擾FENDRR表達(dá),能夠促進(jìn)A549細(xì)胞增殖[(0.85±0.02)vs(0.77±0.08),P0.05]、遷移與侵襲能力(P0.05)。結(jié)論:lnc RNA FENDRR表達(dá)下調(diào)與宣威肺腺癌的發(fā)生發(fā)展相關(guān),其在腫瘤惡性化過(guò)程中抑制腫瘤細(xì)胞的增殖與轉(zhuǎn)移。
[Abstract]:Objective: to detect the expression of non-coding RNA FENDRR long chain in Xuanwei lung adenocarcinoma XWLC-05 cell line and non-small cell lung cancer cell line A549, and to investigate its effect on cell proliferation. Methods: Real-time fluorescence quantitative PCR was used to detect the expression of FENDRR gene in XWLC-05 and A549 cells, and overexpression of p EX-3-FENDRR plasmid was constructed and transfected into XWLC-05 cells, and lentivirus interfered with LV3-FENDRR vector to transfect A549 cells. Real-time fluorescence quantitative PCR was used to detect the expression of FENDRR in XWLC-05 and A549 cells after transfection. The cell proliferation and transwell chamber assay were used to detect the changes of cell migration and invasion ability. Results the expression of FENDRR gene was significantly lower in WXWLC-05 cells than in A549 cells. Compared with the control group, overexpression of FENDRR could significantly inhibit the proliferation of XWLC-05 cells [1.23 鹵0.13 vs 1.46 鹵0.25 p0.05] and decrease its migration and invasion ability, and interfere with the expression of FENDRR. It can promote the proliferation of A549 cell line [0.85 鹵0.02VSD 0.77 鹵0.08P05], and the ability of migration and invasion is P0.05.Conclusion the down-regulation of RNA FENDRR expression is related to the occurrence and development of Xuan Wei lung adenocarcinoma, which inhibits the proliferation and metastasis of tumor cells during the malignant process of tumor.
