自噬基因ARHI與LC3在宮頸癌中的表達(dá)及臨床意義

【摘要】:目的:通過分別檢測并比較在正常宮頸組織、宮頸上皮內(nèi)瘤變(CINII-III)組織、宮頸癌組織中自噬基因ARHI與LC3的表達(dá)水平,并分析兩者與宮頸癌某些臨床病理特征之間的關(guān)系以及兩者之間的相關(guān)性,探討自噬基因ARHI、LC3在宮頸癌發(fā)生、發(fā)展中的作用。方法:隨機(jī)選取2014年11月至2016年10月于青島大學(xué)附屬醫(yī)院嶗山院區(qū)婦科初次收治100例患者的宮頸組織,20例正常宮頸組織、30例宮頸上皮內(nèi)瘤變(CINII-III)組織、50例宮頸癌組織經(jīng)免疫組化染色后ARHI和LC3的表達(dá)水平,并運(yùn)用χ~2檢驗比較各組ARHI、LC3的表達(dá)水平以及兩者與宮頸癌某些臨床病理特征間的關(guān)系,用Spearman法分析宮頸癌中ARHI和LC3之間的相關(guān)性。結(jié)果:1.ARHI在正常宮頸組、宮頸上皮內(nèi)瘤變(CINII-III)組及宮頸癌組中的陽性表達(dá)率依次為:85.0%,73.3%,54.0%,呈下降趨勢,組間差異有統(tǒng)計學(xué)意義(χ~2=7.145,P0.05)。LC3在正常宮頸組、宮頸上皮內(nèi)瘤變(CINII-III)組及宮頸癌組中的陽性表達(dá)率依次為:95.0%,56.7%,38.0%,表達(dá)下調(diào),組間差異有統(tǒng)計學(xué)意義(χ~2=18.801,P0.05)。表明ARHI、LC3與宮頸癌的發(fā)生、發(fā)展密切相關(guān)。2.在宮頸癌組織中,ARHI、LC3的表達(dá)水平與臨床分期及組織分化程度有關(guān),差異有統(tǒng)計學(xué)意義(P0.05),而與淋巴結(jié)是否轉(zhuǎn)移、年齡、SCC無明顯相關(guān)性(P0.05)。3.在宮頸癌組織中,ARHI、LC3的表達(dá)具有正相關(guān)性(r=0.43,p0.05)。4.在上述100例患者中HPV感染呈陽性者共68例,根據(jù)HPV感染程度的不同分為3組,隨著HPV感染程度的加重,ARHI、LC3的表達(dá)水平均呈下降趨勢,但LC3在三組中的組間差異有統(tǒng)計學(xué)意義(χ~2=10.583,P0.05),表明LC3的表達(dá)水平與HPV感染程度密切相關(guān),而ARHI的表達(dá)水平則與HPV感染程度無統(tǒng)計學(xué)意義(P0.05)。結(jié)論:ARHI、LC3表達(dá)水平的降低與缺失可能參與了宮頸癌的發(fā)生、發(fā)展過程,而且兩者具有相關(guān)性,且LC3的表達(dá)水平與HPV感染程度有關(guān)。
[Abstract]:Objective: to detect and compare the expression of autophagy gene ARHI and LC3 in normal cervical tissues, cervical intraepithelial neoplasia (CINII-III) tissues and cervical cancer tissues. The relationship between the two genes and some clinicopathological features of cervical cancer was analyzed, and the relationship between them was analyzed to explore the role of autophagy gene ARHILLC3 in the occurrence of cervical cancer. Methods: from November 2014 to October 2016 in Laoshan Hospital of Qingdao University, 20 cases of normal cervical tissue and 30 cases of cervical intraepithelial neoplasia (CINII-III) were selected randomly. Expression of ARHI and LC3 in 50 cases of cervical carcinoma by immunohistochemical staining. The expression level of ARHILC3 and the relationship between ARHILC3 and some clinicopathological features of cervical cancer were compared by 蠂 ~ 2 test. The correlation between ARHI and LC3 in cervical cancer was analyzed by Spearman method. The positive expression rate of CINII-III in cervical intraepithelial neoplasia group and cervical cancer group was: 85.0% and 73.3% and 54.0% respectively, which showed a downward trend. The difference between the two groups was statistically significant (蠂 ~ 2 / 7.145 ~ (5) P 0.05 / L ~ (3)) in normal cervix. The positive expression rates of CINII-IIII and cervical cancer in the cervical intraepithelial neoplasia group and cervical cancer group were respectively:: 95.00.56.7and 38.0, and the expression was down-regulated. The difference between the two groups was statistically significant (蠂 ~ (2 / 2) 18.801 ~ (18.801)) (P 0.05), which indicated that ARHI LC3 was associated with the occurrence of cervical cancer. The expression level of ARHILC3 was related to the clinical stage and the degree of tissue differentiation. The difference was statistically significant (P 0.05), but the expression level of ARHILC3 was correlated with lymph node metastasis. There was no significant correlation between age and SCC. There was a positive correlation between the expression of ARHILC3 and the expression of ARHILC3 in cervical carcinoma. There were 68 cases with positive HPV infection in the above 100 cases. According to the degree of HPV infection, 68 cases were divided into 3 groups. With the severity of HPV infection, the expression level of ARHILC3 decreased, but the difference of LC3 in the three groups was statistically significant (蠂 ~ (2) 10.583) P 0.05, which indicated that the expression of LC3 was closely related to the degree of HPV infection, but there was a significant difference between the three groups (蠂 ~ (2 +) 10.583), indicating that the expression of LC3 was closely related to the degree of HPV infection. However, there was no significant difference between the expression level of ARHI and the degree of HPV infection. Conclusion the decrease and deletion of the expression level of ARHI LC3 may be involved in the occurrence and development of cervical cancer, and the expression level of LC3 is related to the degree of HPV infection.
