【摘要】：目的 觀察RNF2在乳腺病變組織及乳腺上皮細胞系中的表達,探討其表達與乳腺癌臨床病理特征的關系。方法 運用免疫組化En Vision兩步法檢測21例乳腺良性病變組織和112例乳腺癌組織中RNF2蛋白的表達,分析其表達與乳腺癌臨床病理特征的關系。應用qRT-PCR法檢測2株乳腺正常上皮細胞系和5株乳腺癌細胞系中RNF2基因的表達。結果 乳腺癌組織中RNF2蛋白表達水平與乳腺良性病變組織相比顯著升高(P0.05),且其表達與乳腺癌腫瘤直徑、腋窩淋巴結轉移數目、TNM分期顯著相關(P均0.05);與患者年齡、組織學分級、ER、PR、HER-2表達情況無顯著相關(P均0.05);乳腺癌細胞系RNF2 mRNA水平亦高于乳腺良性上皮細胞系(P0.05)。結論 RNF2在乳腺癌組織中高表達,可作為乳腺癌治療的潛在靶點。
[Abstract]:Objective To observe the expression of RNF2 in breast carcinoma tissue and mammary epithelial cell lines, to investigate the relationship between the expression and clinicopathological features of breast cancer. The expression of RNF2 protein immunohistochemistry method using En Vision two step method was used to detect 21 cases of benign breast lesions and 112 cases of breast cancer and the relationship between their expression and clinical pathological features breast cancer. The expression of qRT-PCR was used to detect 2 strains of normal breast epithelial cells and 5 strains of RNF2 gene in breast cancer cell lines. The RNF2 protein in breast carcinoma and expression of benign breast lesions were significantly higher than (P0.05), and the expression of breast cancer and tumor diameter, number of metastatic lymph nodes. TNM stage were significantly correlated (P 0.05); and the patients age, histological grade, ER, PR, HER-2 expression had no significant correlation (P 0.05); breast cancer cell line RNF2 mRNA level is also higher than that in benign breast epithelium Cell line (P0.05). Conclusion RNF2 is highly expressed in breast cancer tissue and can be used as a potential target for the treatment of breast cancer.
