【摘要】：目的:研究飛燕草素(delphinidin,Dp)聯(lián)合自噬抑制劑3-甲基腺嘌呤(3-Methyladnine,3-MA)對(duì)HER-2+乳腺癌細(xì)胞MDA-MB-453的抑制效應(yīng)。方法:用不同濃度飛燕草素處理MDA-MB-453細(xì)胞,CCK-8檢測細(xì)胞增殖情況;熒光斑點(diǎn)實(shí)驗(yàn)和Weston Blot實(shí)驗(yàn)檢測飛燕草素對(duì)細(xì)胞自噬的誘導(dǎo)情況;Weston Blot實(shí)驗(yàn)檢測3-MA對(duì)自噬的抑制狀況,CCK-8檢測飛燕草素聯(lián)合3-MA對(duì)細(xì)胞增殖狀況的影響。結(jié)果:飛燕草素能有效抑制MDA-MB-453細(xì)胞增殖,誘導(dǎo)MDA-MB-453細(xì)胞自噬并呈劑量依賴效應(yīng);3-MA有效抑制MDA-MB-453細(xì)胞自噬,同時(shí)增強(qiáng)自噬飛燕草素抗HER-2+乳腺癌細(xì)胞MDA-MB-453增殖效應(yīng)。結(jié)論:飛燕草素聯(lián)合自噬抑制劑3-MA,能促進(jìn)飛燕草素抗HER-2+乳腺癌細(xì)胞MDA-MB-453增殖。
[Abstract]:Objective: to study the inhibitory effect of Delphinidin (Dp) and autophagy inhibitor 3-Methyladenine (3-Methyladenine 3-MA) on MDA-MB-453 of HER-2 breast cancer cells. Methods: the proliferation of MDA-MB-453 cells treated with different concentrations of Delphinidin was assayed by CCK-8. Fluorescence speckle assay and Weston Blot assay were used to detect the induction of autophagy induced by gliadin and the inhibitory effect of 3-MA on autophagy was detected by Weston Blot assay. Results: the proliferation of cells was determined by CCK-8 combined with 3-MA. Can effectively inhibit the proliferation of MDA-MB-453 cells, MDA-MB-453 cells were induced to autophagy and inhibited the autophagy of MDA-MB-453 cells in a dose-dependent manner. Conclusion: combined with autophagy inhibitor 3-MA, it can promote the proliferation of HER-2 breast cancer cell line MDA-MB-453.
