肺腺癌中缺氧誘導因子-1α調控程序性死亡因子配體1的表達

【摘要】:目的研究缺氧誘導因子-1α(HIF-1α)和程序性死亡因子配體1(PD-L1)在肺腺癌中的表達與臨床病理特征的關系,探討肺腺癌中HIF-1α對PD-L1的調控作用。方法應用免疫組織化學方法檢測99例肺腺癌組織中HIF-1α、PD-L1的表達。Co Cl2模擬化學性缺氧處理肺腺癌細胞,并將其分為正常組及CoCl_2組,qRT-PCR檢測HIF-1α、PD-L1 mRNA表達,Western blotting檢測HIF-1α、PD-L1蛋白表達。siRNA-HIF-1α瞬時轉染肺腺癌細胞,并將其分為陰性對照組及siR-HIF-1α組,qRT-PCR、Western blotting檢測轉染效率及PD-L1 mRNA和蛋白的表達。結果 99例肺腺癌組織中,HIF-1α和PD-L1的陽性率分別為47.48%和34.34%,HIF-1α表達水平與患者年齡和臨床分期有關(P0.05),PD-L1表達水平與患者臨床分期有關(P0.05),HIF-1α蛋白表達與PD-L1蛋白表達呈正相關(P0.01)。與正常組相比,CoCl_2組細胞中HIF-1α蛋白表達水平明顯升高(t=-2.394,P=0.029),PD-L1 mRNA及蛋白表達水平隨之明顯升高(t=-3.146,P=0.006;t=-3.308,P=0.004)。與陰性對照組相比,siR-HIF-1α組沉默處理細胞后,PD-L1 mRNA及蛋白表達水平顯著降低(t=2.746,P=0.014;t=2.339,P=0.033)。結論肺腺癌組織中HIF-1α和PD-L1明顯共表達;PD-L1表達受HIF-1α調控。
[Abstract]:Objective to study the relationship between the expression of hypoxia inducible factor-1 偽 (HIF-1 偽) and programmed death factor ligands 1 (PD-L1) in lung adenocarcinoma and the clinicopathological features. To investigate the regulatory effect of HIF-1 偽 on PD-L1 in lung adenocarcinoma. Methods the expression of HIF-1 偽 PD-L1 in 99 cases of lung adenocarcinoma was detected by immunohistochemical method. The expression of PD-L1 in 99 cases of lung adenocarcinoma was treated with simulated chemical hypoxia. It was divided into two groups: normal group and CoCl_2 group. The expression of HIF-1 偽 -PD-L1 mRNA was detected by qRT-PCR. Western blotting was used to detect the expression of HIF-1 偽 -pPD-L1 protein. SiRNA-HIF-1 偽 was transiently transfected into lung adenocarcinoma cells. The efficiency of transfection and the expression of PD-L1 mRNA and protein in 99 cases of lung adenocarcinoma were detected by Western blotting. Results the positive rates of HIF-1 偽 and PD-L1 in 99 cases of lung adenocarcinoma were 47.48% and 34.34%, respectively, and the expression levels of HIF-1 偽 were correlated with the age and clinical stage of the patients. There was a positive correlation between the expression of P0.05HIF-1 偽 protein and the expression of PD-L1 protein. Compared with the normal group, the expression level of HIF-1 偽 protein in the cells of CoCl2 group was significantly higher than that in the normal group. The expression level of PD-L1 mRNA and protein increased significantly. Compared with control group, the expression of PD-L1 mRNA and protein in the silenced cells of the negative control group was significantly lower than that in the control group. Conclusion the expression of HIF-1 偽 and PD-L1 in lung adenocarcinoma is regulated by HIF-1 偽.
