HER2和TSPAN1蛋白在胃癌組織中的表達及臨床病理意義

【摘要】:目的:(1)探討HER2和TSPAN1蛋白在胃癌發(fā)生發(fā)展中的作用及意義。(2)探討HER2和TSPAN1蛋白的表達與胃癌患者年齡、性別、腫瘤大小、部位、分化程度、轉(zhuǎn)移、Lauren分型和TNM分期的關(guān)系及意義。(3)探討HER2和TSPAN1蛋白在胃癌發(fā)展中是否有協(xié)同關(guān)系。方法:采用Envision免疫組織化學(xué)法(二步法)對64例胃癌組織和癌旁粘膜組織進行HER2和TSPAN1蛋白表達的檢測,同時選取20例正常胃粘膜組織作為對照。結(jié)果:(1)64例胃癌組織中HER2蛋白陽性表達率23.4%(15/64)顯著高于20例正常胃粘膜組織0%(0/20),經(jīng)統(tǒng)計學(xué)分析,差異具有顯著性(P0.05);64例癌旁粘膜組織中HER2蛋白陽性表達率10.9%(7/64)高于20例正常胃粘膜組織0%(0/20),經(jīng)統(tǒng)計學(xué)分析,差異無顯著性(P0.05);64例胃癌組織中HER2蛋白陽性表達率23.4%(15/64)高于64例癌旁粘膜組織10.9%(7/64),經(jīng)統(tǒng)計學(xué)分析,差異無顯著性(P0.05)。(2)Lauren分型42例腸型胃癌組織中HER2蛋白陽性表達率33.3%(14/42)顯著高于22例彌漫型及混合型胃癌組織4.5%(1/22),經(jīng)統(tǒng)計學(xué)分析,差異具有顯著性(P0.05)。13例高分化胃癌組織38.5%(5/13)、29例中分化胃癌組織中HER2蛋白陽性表達率31.0%(9/29)顯著高于22例低分化胃癌組織4.5%(1/22),經(jīng)統(tǒng)計學(xué)分析,差異具有顯著性(P0.05)。38例淋巴結(jié)轉(zhuǎn)移胃癌組織中HER2蛋白陽性表達率36.8%(14/38)顯著高于26例無淋巴結(jié)轉(zhuǎn)移胃癌組織3.8%(1/26),經(jīng)統(tǒng)計學(xué)分析,差異具有顯著性(P0.05)。HER2蛋白陽性表達與患者年齡、性別、腫瘤大小、部位、TNM分期無關(guān)(P0.05)。(3)64例胃癌組織中TSPAN1蛋白陽性表達率68.8%(44/64)顯著高于20例正常胃粘膜組織0%(0/20),經(jīng)統(tǒng)計學(xué)分析,差異具有顯著性(P0.05);64例癌旁粘膜組織中TSPAN1蛋白陽性表達率23.4%(15/64)顯著高于20例正常胃粘膜組織0%(0/20),經(jīng)統(tǒng)計學(xué)分析,差異有顯著性(P0.05);64例胃癌組織中TSPAN1蛋白陽性表達率68.8%(44/64)顯著高于64例癌旁粘膜組織23.4%(15/64),經(jīng)統(tǒng)計學(xué)分析,差異具有顯著性(P0.05)。(4)Lauren分型42例腸型胃癌組織中TSPAN1蛋白陽性表達率52.4%(22/42)顯著低于22例彌漫型及混合型胃癌組織100%(22/22),經(jīng)統(tǒng)計學(xué)分析,差異具有顯著性(P0.05)。22例低分化胃癌組織100%(22/22)、29例中分化胃癌組織中TSPAN1蛋白陽性表達率62.1%(18/29)顯著高于13例高分化胃癌組織30.8%(4/13),經(jīng)統(tǒng)計學(xué)分析,差異具有顯著性(P0.05)。TSPAN1蛋白陽性表達與患者年齡、性別、腫瘤大小、部位、轉(zhuǎn)移、TNM分期無關(guān)。(5)胃癌組織中HER2和TSPAN1蛋白表達有一定的相關(guān)性(X2=7.107,r=0.349,P=0.003)。結(jié)論:(1)HER2和TSPAN1蛋白陽性表達與胃癌的發(fā)生發(fā)展有關(guān)。(2)HER2和TSPAN1蛋白陽性表達與胃癌分化程度及Lauren分型有關(guān),而且HER2蛋白陽性表達與胃癌患者的轉(zhuǎn)移也有關(guān)。(3)HER2和TSPAN1蛋白陽性表達與胃癌患者年齡、性別、腫瘤大小、部位、TNM分期無關(guān),TSPAN1蛋白陽性表達與轉(zhuǎn)移無關(guān)。
[Abstract]:Objective to explore the role and significance of HER2 and TSPAN1 proteins in the development of gastric cancer. (2) to explore the relationship between the expression of HER2 and TSPAN1 proteins and the age, sex, tumor size, location and differentiation of gastric cancer patients. The relationship between TNM staging and HER2 classification) to explore whether there is a synergistic relationship between HER2 and TSPAN1 protein in the development of gastric cancer. Methods: 64 gastric cancer tissues and adjacent mucosa tissues were studied by Envision immunohistochemical method. HER2 and TSPAN1 protein expression were detected. At the same time, 20 normal gastric mucosa tissues were selected as control. Results the positive expression rate of HER2 protein in 64 cases of gastric cancer was significantly higher than that of 20 cases of normal gastric mucosa tissue, and the positive rate of HER2 protein in 64 cases of gastric cancer was significantly higher than that of 20 cases of normal gastric mucosa tissue. The positive expression rate of HER2 protein in 64 cases of paracancerous mucosa was significantly higher than that of 20 cases of normal gastric mucosa (10.9% / 64), and was significantly higher than that of 20 cases of normal gastric mucosa tissue (0 / 20%). The positive expression rate of HER2 protein in 64 cases of gastric cancer was 23.4% (15 / 64) higher than that of 10.9% (10.9%) in 64 cases of gastric cancer. The positive expression rate of HER2 protein in 42 cases of intestinal type gastric carcinoma was significantly higher than that of 22 cases of diffuse type and mixed type of gastric cancer. The positive expression rate of HER2 protein in 42 cases of intestinal type gastric carcinoma was significantly higher than that of 22 cases of diffuse type and mixed type of gastric carcinoma. The positive expression rate of HER2 protein in 29 cases of well-differentiated gastric carcinoma was significantly higher than that in 22 cases of poorly differentiated gastric carcinoma (P 0.05). The expression rate of HER2 protein in 29 cases of well-differentiated gastric carcinoma was significantly higher than that of 22 cases of poorly differentiated gastric carcinoma, and the expression rate of HER2 protein was significantly higher than that in 22 cases of poorly differentiated gastric carcinoma, and the expression rate of HER2 protein in 29 cases was significantly higher than that in 22 cases of poorly differentiated gastric carcinoma. The positive expression rate of HER2 protein in 38 cases with lymph node metastasis was significantly higher than that in 26 cases without lymph node metastasis. The positive expression rate of HER2 protein in 38 cases of lymph node metastasis was significantly higher than that of 26 cases without lymph node metastasis. By statistical analysis, the positive expression of HER2 protein in patients with lymph node metastasis was significantly higher than that in patients with age and sex. The positive expression rate of TSPAN1 protein in 64 cases of gastric cancer was significantly higher than that in 20 cases of normal gastric mucosa. The positive expression rate of TSPAN1 protein in 64 paracancerous mucosa tissues was significantly higher than that in 20 normal gastric mucosa tissues. The positive expression rate of TSPAN1 protein in 64 cases of gastric cancer was significantly higher than that in the adjacent mucosa of 64 cases (68.8% / 64%), which was significantly higher than that in the adjacent mucosa tissue of 64 cases (23.4% / 64%). The positive expression rate of TSPAN1 protein in 42 cases of intestinal type gastric carcinoma was significantly lower than that in 22 cases of diffuse type and mixed type of gastric cancer. The expression rate of TSPAN1 protein in 42 cases of intestinal type gastric carcinoma was significantly lower than that of 22 cases of diffuse type and mixed type of gastric carcinoma. The positive expression rate of TSPAN1 protein in 22 cases of poorly differentiated gastric carcinoma was significantly higher than that in 13 cases of well-differentiated gastric carcinoma. The positive expression rate of TSPAN1 protein was significantly higher than that in 13 cases of well-differentiated gastric carcinoma. The difference was statistically significant between P0.05. TSPAN1 protein expression and age. Sex, tumor size, location, There was a correlation between the expression of HER2 and TSPAN1 protein in gastric cancer tissues. Conclusion the positive expression of HER2 and TSPAN1 proteins is related to the occurrence and development of gastric carcinoma, and the expression of HER2 and TSPAN1 protein is related to the differentiation of gastric carcinoma and the type of Lauren. Moreover, the positive expression of HER2 protein was related to metastasis of gastric cancer patients. The positive expression of HER2 and TSPAN1 protein was not related to age, sex, tumor size and tumor stage of gastric cancer patients. There was no correlation between the positive expression of TSPAN1 protein and metastasis.
