【摘要】：目的探討Numb和GPC3在原發(fā)性肝細(xì)胞肝癌中的表達(dá)及二者的相關(guān)性,探究Numb和GPC3在原發(fā)性肝細(xì)胞肝癌中的臨床價值。方法選擇鄭州大學(xué)第一附屬醫(yī)院肝膽胰外科于2008年6月至2011年6月收治的原發(fā)性肝細(xì)胞肝癌患者共60例,同時選取27例相同時間段的肝血管瘤及肝外傷的患者。分別統(tǒng)計分析,Numb與GPC3在原發(fā)性肝細(xì)胞肝癌組織、癌旁組織、正常組織中的表達(dá)情況,同時對比Numb和GPC3陽性表達(dá)率與各組患者臨床資料的關(guān)系,以及Numb和GPC3在三種組織中的表達(dá)相關(guān)性。結(jié)果原發(fā)性肝細(xì)胞肝癌組織中Numb蛋白主要呈現(xiàn)陰性,癌旁組織中Numb蛋白主要呈現(xiàn)陰性或弱陽性,正常肝組織中主要呈現(xiàn)弱陽性或陽性,三組間陽性表達(dá)率差異有統(tǒng)計學(xué)意義(PO.05)。對比臨床資料,Numb蛋白表達(dá)水平與三組患者的肝硬化程度、腫瘤大小,年齡及性別無關(guān),差異無統(tǒng)計學(xué)意義(P0.05),而與患者Edmondson分級及腫瘤包膜、TNM分期及門脈癌栓有關(guān),差異均有統(tǒng)計學(xué)意義(P0.05)。即Edmondson分級III~Ⅳ級、TNM分期Ⅲ期、無腫瘤包膜但有門脈癌栓的患者,Numb蛋白在三組患者中主要呈現(xiàn)陰性或弱陽性,三組患者之間陽性表達(dá)率差異具有統(tǒng)計學(xué)意義(P0.05)。同時,相對于正常肝臟組織來說原發(fā)性肝細(xì)胞肝癌組織中GPC3的陽性表達(dá)率則大大升高,差異具有統(tǒng)計學(xué)意義(P0.05)�；颊叩母斡不潭取⒛[瘤大小、年齡、性別則與GPC3的表達(dá)水平無關(guān),差異無統(tǒng)計學(xué)意義(P0.05)。而與患者Edmondson分級及腫瘤包膜、TNM分期及門脈癌栓有關(guān),差異均有統(tǒng)計學(xué)意義(P0.05)。即對于Edmondson分級處于Ⅲ~Ⅳ級及無腫瘤包膜、TNM分期處于III期及存在門脈癌栓的患者,GPC3陽性率將明顯提高,差異有統(tǒng)計學(xué)意義(P0.05)。GPC3蛋白表達(dá)與Numb表達(dá)呈負(fù)相關(guān)(P0.05)。結(jié)論原發(fā)性肝細(xì)胞肝癌的侵襲性與Numb蛋白陽性表達(dá)率下降及GPC3陽性表達(dá)率上升均相關(guān),且二者負(fù)相關(guān)表達(dá)。因而對原發(fā)性肝細(xì)胞肝癌患者來說,Numb蛋白與GPC3的表達(dá)對其預(yù)后的判定可能具有一定的臨床指導(dǎo)作用。
[Abstract]:Objective to investigate the expression of Numb and GPC3 in hepatocellular carcinoma (HCC) and their correlation. To explore the clinical value of Numb and GPC3 in primary hepatocellular carcinoma (HCC). Methods Primary hepatic fine liver was selected from June 2008 to June 2011 in the Department of Hepatobiliary and Pancreatic surgery, the first affiliated Hospital of Zhengzhou University. There were 60 patients with hepatocellular carcinoma. At the same time, 27 patients with hepatic hemangioma and liver trauma were selected to analyze the expression of GPC3 and numb in primary hepatocellular carcinoma (HCC), paracancerous tissues and normal tissues respectively. At the same time, the relationship between the positive expression rate of Numb and GPC3 and the clinical data of each group was compared. The expression of Numb and GPC3 in three tissues were correlated. Results Numb protein was negative in primary hepatocellular carcinoma (HCC). Numb protein was mainly negative or weakly positive in paracancerous tissues and weakly positive or positive in normal liver tissues. The difference of positive expression rate among the three groups was statistically significant (P < 0.05). The clinical data were compared. The expression of Numb protein was not related to the degree of cirrhosis, tumor size, age and sex, but there was no significant difference between the three groups (P 0.05). However, there were significant differences in Edmondson grade, tumor capsule TNM stage and portal vein tumor thrombus. The difference was significant (P 0.05), that is, Edmondson grade III- 鈪，
