【摘要】:目的:探討WW結(jié)構(gòu)域E3泛連接酶1(WWP1)在乳腺癌病人組織中的表達(dá),分析與乳腺癌病人臨床病理因素的關(guān)系,并討論WWPl基因和乳腺癌患者中ER、PR、P53基因、HER-2、年齡、淋巴結(jié)轉(zhuǎn)移、病理組織學(xué)分級(jí)以及腫瘤組織大小之間的關(guān)系,為乳腺癌病人的臨床治療及預(yù)后提供科學(xué)依據(jù)。材料與方法:該實(shí)驗(yàn)選取2004年3月一2007年6月接受乳腺癌切除術(shù)的129例女性患者乳腺癌組織存檔石蠟標(biāo)本,應(yīng)用組織芯片技術(shù)來(lái)構(gòu)建乳腺癌病人組織芯片,通過(guò)HE染色和免疫組化染色來(lái)判斷組織芯片質(zhì)量以及觀察WWP1表達(dá)情況;另選取乳腺良性病人手術(shù)切除的19例患者乳腺組織存檔的石蠟標(biāo)本玻片來(lái)作為對(duì)照組。從而將乳腺癌病人的年齡大小、腫瘤組織的大小、腋窩淋巴結(jié)侵襲轉(zhuǎn)移、臨床病理分期以及乳腺癌臨床指標(biāo)ER、PR和HER-2基因的表達(dá)情況與WWP1表達(dá)情況作相關(guān)分析,并得出結(jié)論。結(jié)果:HE玻片染色顯示兩組組織芯片染色合格,可以代表判斷乳腺癌組織病理學(xué)形態(tài)。通過(guò)免疫組化染色觀察得到,在乳腺癌病人組織中有92例(約70.5%)可見(jiàn)WWP1基因表達(dá)為陽(yáng)性,乳腺良性組織中均未見(jiàn)WWP1表達(dá)。術(shù)后有腋窩淋巴結(jié)轉(zhuǎn)移的病人WWP1陽(yáng)性表達(dá)率明顯高于WWP1陰性表達(dá)者(P0.05);HER-2基因的表達(dá)和WWP1的表達(dá)呈正相關(guān)(P0.05);腫瘤大小與WWP1陽(yáng)性表達(dá)率呈正相關(guān)(P0.05);乳腺癌組織的腫瘤分期和WWP1的陽(yáng)性表達(dá)率之間無(wú)明顯統(tǒng)計(jì)學(xué)意義(P0.05)。雌激素受體、孕激素受體表達(dá)與WWPl陽(yáng)性表達(dá)率比較差異均無(wú)統(tǒng)計(jì)學(xué)意義(P0.05)。WWP1的表達(dá)和患者生存之間沒(méi)有明顯統(tǒng)計(jì)學(xué)意義(P0.05)。結(jié)論:WWP1和雌激素受體(ER)、孕激素受體(PR)、腫瘤病理分期之間無(wú)明顯統(tǒng)計(jì)學(xué)意義,且WWP1和患者的預(yù)后生存之間無(wú)明顯統(tǒng)計(jì)學(xué)意義;WWP1在乳腺癌組織中過(guò)表達(dá)且與腫瘤組織大小、淋巴結(jié)轉(zhuǎn)移及Her-2基因的表達(dá)正相關(guān),WWP1有望成為一個(gè)新的治療靶點(diǎn)。
[Abstract]:Objective: To investigate the WW domain of E3 (WWP1) - ligase 1 expression in tissue of breast cancer patients in the relationship analysis and clinical pathological factors in breast cancer patients, and discuss the WWPl gene in patients with breast cancer and ER, PR, P53, HER-2 gene, age, lymph node metastasis, histological grade and the relationship between the size of the tumor tissue, and provide scientific basis for clinical treatment and prognosis of patients with breast cancer. Materials and methods: the experiment selected in March 2004 June 2007 to accept 129 female patients with breast cancer paraffin samples of resection of breast cancer, the application of tissue microarray technology to construct breast cancer patients by tissue chip, HE staining and immunohistochemical staining to determine the expression of chip quality and observation WWP1; the other from 19 cases of breast excision of benign breast tissues of patients with archived paraffin samples of patients as control group were. The correlation between the age of breast cancer, the size of tumor tissue, the invasion and metastasis of axillary lymph nodes, the clinicopathological staging, and the expression of ER, PR and HER-2 genes in breast cancer, and the expression of WWP1 were analyzed. Results: the staining of HE slides showed that two groups of tissue chips were coloring, which could represent the histopathological morphology of breast cancer. By immunohistochemical staining, 92 cases (70.5%) were found in breast cancer patients, and the expression of WWP1 gene was positive. There was no WWP1 expression in benign breast tissues. After the operation was significantly higher than the negative expression of WWP1 in patients with positive expression of WWP1 metastasis of axillary lymph node (P0.05); their expression is positively correlated with the expression of WWP1 and the HER-2 gene (P0.05) was positively correlated with tumor size; the positive expression rate of WWP1 (P0.05); breast cancer tissue and tumor stage between the positive expression rate of WWP1 no statistical significance (P0.05). There was no significant difference between the expression of estrogen receptor and progesterone receptor and the positive expression rate of WWPl (P0.05). There was no significant statistical significance between the expression of WWP1 and the survival of the patients (P0.05). Conclusion: WWP1 and estrogen receptor (ER) and progesterone receptor (PR), tumor pathological stage was not statistically significant, and prognosis in patients with WWP1 and no statistical significance; in the WWP1 over expression in breast cancer tissue and the expression and tumor size, lymph node metastasis and Her-2 gene positive correlation, WWP1 is expected to become a new therapeutic target.
