【摘要】：目的:檢測(cè)上皮間質(zhì)轉(zhuǎn)化(epithelial-mesenchymal transition,EMT)標(biāo)記蛋白及β-連環(huán)素(β-catenin)、糖原合成酶激酶-3β(Glycogen synthase kinase3β,GSK-3β)在口腔鱗狀細(xì)胞癌(Oral squamous cell carcinoma,OSCC)組織的表達(dá),初步探索其在OSCC發(fā)生EMT中的作用。材料與方法:通過免疫組織化學(xué)(Immunohistochemistry,IHC)的方法檢測(cè)61例OSCC組織和13例正常組織中上皮源性標(biāo)記物:E-cadherin、間質(zhì)來源標(biāo)記物:N-cadherin和β-catenin、GSK-3β、Snail、p-GSK-3β的表達(dá)情況,并與臨床病理資料進(jìn)行對(duì)照、分析。結(jié)果:β-catenin、N-cadherin、Snail、pGSK-3β在OSCC組的表達(dá)明顯高于正常組,GSK-3β及E-cadherin的表達(dá)明顯低于正常組織組(P0.05);β-catenin、pGSK-3β、Snai、N-cadherin的在有淋巴結(jié)轉(zhuǎn)移組中表達(dá)高于無淋巴結(jié)轉(zhuǎn)移組,E-cadherin、GSK-3β的表達(dá)低于無淋巴結(jié)轉(zhuǎn)移組(P0.05)。β-catenin、E-cadherin、GSK-3β、N-cadherin、Snail在OSCC中表達(dá)兩兩相關(guān)(P0.05)。E-cadherin(-),N-cadherin(+)蛋白陽性表型患者在中低分化組較高分化組多、在臨床III+IV分期患者較臨床I+II分期患者多,在有淋巴轉(zhuǎn)移組較無淋巴轉(zhuǎn)移組多(P0.05)。E-cadherin(-)、N-cadherin(+)蛋白表型組與β-catenin、pGSK-3β表達(dá)呈正相關(guān)(r1=0.411,r2=0.320,P0.05),E-cadherin(-)、N-cadherin(+)蛋白表型組與GSK-3β表達(dá)呈負(fù)相關(guān)(r=-0.341,P=0.07)。E-cadherin(-)、N-cadherin(+)、β-catenin(+)、pGSK-3β(+)蛋白陽性表型患者在中低分化組較高分化組多、在臨床III+IV期患者較臨床I+II分期患者多,在有淋巴轉(zhuǎn)移組較無淋巴轉(zhuǎn)移組多(P0.05)。結(jié)論:1、OSCC中存在上皮間質(zhì)轉(zhuǎn)化現(xiàn)象。2、GSK-3β、β-catenin可能在OSCC組織發(fā)生上皮間質(zhì)轉(zhuǎn)化現(xiàn)象的過程中起一定作用,聯(lián)合上述因子共同檢測(cè),可能對(duì)于預(yù)判OSCC是否發(fā)生轉(zhuǎn)移及針對(duì)口腔鱗癌的靶向治療有一定幫助。
