PM2.5對(duì)MLE-12細(xì)胞的損傷及水通道蛋白5mRNA表達(dá)的影響

【摘要】:目的:研究大氣細(xì)顆粒物(PM 2.5)對(duì)體外培養(yǎng)的肺泡Ⅱ型上皮細(xì)胞系MLE-12細(xì)胞活性、炎性因子分泌及其水通道蛋白5mRNA表達(dá)的影響,探討PM 2.5對(duì)呼吸系統(tǒng)的損傷及其機(jī)制。方法:采集大氣中的PM2.5,并制成懸浮液,MLE-12細(xì)胞暴露于不同濃度的PM2.5處,用MTT比色法測(cè)定細(xì)胞存活力,放射免疫法測(cè)定炎性因子TNF-α、IL-6和IL-8的含量,熒光實(shí)時(shí)定量PCR法檢測(cè)AQP5 mRNA的表達(dá)。結(jié)果:MTT實(shí)驗(yàn)中不同濃度的PM2.5作用于MLE-12細(xì)胞24 h、48 h后,細(xì)胞存活率顯著下降,并呈濃度效應(yīng)關(guān)系。炎性因子測(cè)定中,隨著PM2.5濃度的增加,PM2.5誘導(dǎo)MLE-12細(xì)胞產(chǎn)生炎性因子TNF-α、IL-6和IL-8的濃度逐漸升高(P0.05)。在0.025~0.20 mg/m L的質(zhì)量濃度范圍內(nèi),隨著PM2.5染毒液濃度的增加,MLE-12中AQP5mRNA表達(dá)水平逐漸降低,且與對(duì)照組相比,差異有統(tǒng)計(jì)學(xué)意義(P0.05)。結(jié)論:PM2.5通過誘導(dǎo)MLE-12細(xì)胞的炎癥反應(yīng)及AQP5蛋白表達(dá)降低引起MLE-12細(xì)胞的活性降低,促進(jìn)細(xì)胞凋亡。
[Abstract]:Objective: to study the effects of atmospheric fine particles (PM 2.5) on the activity, inflammatory factor secretion and aquaporin 5mRNA expression of alveolar type 2 epithelial cell line MLE-12 in vitro. To investigate the injury of respiratory system induced by PM 2.5 and its mechanism. Methods: PM2.5, in the atmosphere was collected and made into suspension. MLE-12 cells were exposed to different concentrations of PM2.5. The cell viability was measured by MTT colorimetric assay, and the contents of inflammatory factors TNF- 偽, IL-6 and IL-8 were measured by radioimmunoassay. The expression of AQP5 mRNA was detected by fluorescence real-time quantitative PCR. Results: MLE-12 cells were treated with different concentrations of PM2.5 for 24 h in MTT assay, and the survival rate of MLE-12 cells decreased significantly at 48 h, in a concentration-dependent manner. In the determination of inflammatory factors, with the increase of PM2.5 concentration, the concentration of inflammatory factors TNF- 偽, IL-6 and IL-8 induced by PM2.5 increased gradually (P 0.05). In the range of 0.025 mg/m / L, with the increase of PM2.5 concentration, the expression of AQP5mRNA in MLE-12 decreased gradually, and the difference was statistically significant compared with the control group (P 0.05). Conclusion: PM2.5 can induce inflammatory reaction of MLE-12 cells and decrease the expression of AQP5 protein to decrease the activity of MLE-12 cells and promote apoptosis.
