亞砷酸鈉對Hacat細胞GSK-3β及CyclinD1表達的影響

【摘要】:目的探討亞砷酸鈉對人上皮角質(zhì)形成細胞系(Hacat細胞)PI3K/AKT信號通路中細胞周期相關(guān)分子GSK-3β、CyclinD1的影響。方法將人Hacat細胞暴露于濃度分別為0、10、25、50、100μmol/L的亞砷酸鈉6 h,采用Western Blot法檢測細胞總蛋白的GSK-3β,磷酸化GSK-3β(p-GSK-3β),CyclinD1和磷酸化CyclinD1(p-CyclinD1)的蛋白表達水平。結(jié)果與對照組比較,10、25、50、100μmol/L染砷組總蛋白中p-GSK-3β和CyclinD1蛋白表達水平均顯著增高(P0.05),而p-CyclinD1蛋白表達水平顯著降低(P0.05)。結(jié)論亞砷酸鈉可影響Hacat細胞中的GSK-3β分子,使其磷酸化失活,進而失去對細胞周期相關(guān)蛋白CyclinD1的磷酸化作用,從而促進CyclinD1的表達。
[Abstract]:Aim to investigate the effect of sodium arsenite on GSK-3 尾 and CyclinD1 in PI3K/AKT signaling pathway of human epithelial keratinocytes (Hacat cells). Methods Human Hacat cells were exposed to sodium arsenite at concentrations of 0, 10, 25 and 50 100 渭 mol / L for 6 h, respectively. The expression levels of GSK-3 尾, phosphorylated GSK-3 尾 (p-GSK-3 尾), CyclinD1 and phosphorylated CyclinD1 (p-CyclinD1) were detected by Western Blot assay. Results compared with the control group, the expression levels of p-GSK-3 尾 and CyclinD1 protein in the total protein of 10,25,50100 渭 mol / L arsenic exposed group were significantly increased (P0.05), while the expression level of p-CyclinD1 protein was significantly decreased (P0.05). Conclusion Sodium arsenite can affect the GSK-3 尾 molecule in Hacat cells, make it phosphorylated and inactivated, and then lose the phosphorylation of CyclinD1, thus promote the expression of CyclinD1.
