嬰兒期腸道菌群結(jié)構(gòu)改變規(guī)律的初探

【摘要】:目的分析嬰兒期腸道菌群結(jié)構(gòu)的動(dòng)態(tài)變化,探討嬰兒腸道菌群定植演替的一般規(guī)律。方法以正常足月新生兒為研究對(duì)象,采用PCR-DGGE技術(shù)檢測(cè)嬰兒出生后1、2、4、7、10、14、28 d,3、6、12月這10個(gè)時(shí)間點(diǎn)糞便中菌群結(jié)構(gòu)。結(jié)果嬰兒腸道內(nèi)細(xì)菌豐度及Shannon-Wiener多樣性指數(shù)出生后d 1~d 10出現(xiàn)上升,隨后轉(zhuǎn)為下降,至d 28達(dá)到最低值,隨后再次上升,12月時(shí)達(dá)到峰值。相似性及聚類(lèi)分析顯示,嬰兒腸道菌群結(jié)構(gòu)d 2~d 28聚為一類(lèi),d 1、3~12月聚為一類(lèi),但d 1與其他時(shí)間點(diǎn)相似性較低。結(jié)論嬰兒出生后d 1腸道內(nèi)即有細(xì)菌開(kāi)始定植,d 2~d28菌群結(jié)構(gòu)發(fā)生顯著改變,但尚處于較為平穩(wěn)狀態(tài);3個(gè)月后腸道內(nèi)細(xì)菌定植速度加快。
[Abstract]:Objective to analyze the dynamic changes of intestinal flora in infancy and to explore the general rules of colonization succession of intestinal flora in infants. Methods PCR-DGGE technique was used to detect the fecal flora of normal full-term newborns. Results the bacterial abundance and Shannon-Wiener diversity index in the intestinal tract of infants increased after birth, then decreased to the lowest level on d28, then increased again, and reached the peak at 12 months. The results of similarity and cluster analysis showed that the intestinal flora structure of infants was divided into two groups: d2D28 and d1 ~ 3 ~ 12, but the similarity between D1 and other time points was low. Conclusion on the first day after birth, bacteria began to colonize the intestinal tract, and the structure of d 2~d28 flora changed significantly, but it was still in a stable state, and the colonization rate of the bacteria in the intestinal tract was accelerated after 3 months.
