實時熒光PCR法同時檢測食物中大豆和芹菜致敏原成分

【摘要】:大豆、芹菜等食品原料是一些特殊人群的致敏原。根據(jù)大豆atp A基因和芹菜mtd基因設計特異性引物,利用不同熒光素標記的Taq Man探針,建立了一種實時熒光PCR檢測方法,可同時檢測食物中大豆和芹菜致敏原成分。分別以大豆、芹菜、大米、小麥、大麥、花生、芝麻、玉米、馬鈴薯、蕃茄、核桃、開心果、腰果、葵花籽、杏仁、蘋果、梨、草莓、豬肉、牛肉、羊肉及魚肉等材料的基因組DNA作為模板,進行PCR擴增實驗,結果發(fā)現(xiàn)該方法僅能特異性擴增大豆和芹菜致敏原成分。靈敏度測試結果表明大豆和芹菜成分的檢出限均達0.01%。因此,本方法可以作為同時檢測食品中大豆和芹菜致敏原成分的特異性方法。
[Abstract]:Soybean, celery and other food raw materials are some special people allergen. Based on the design of specific primers of soybean atp A gene and celery mtd gene, a real-time fluorescent PCR detection method was developed using Taq Man probes labeled with different fluorescein, which can be used to detect the allergen components of soybean and celery simultaneously. With soybeans, celery, rice, wheat, barley, peanuts, sesame, corn, potatoes, tomatoes, walnuts, pistachios, cashews, sunflower seeds, almonds, apples, pears, strawberries, pork, beef, The genomic DNA of mutton and fish was used as template for PCR amplification. The results showed that this method could only amplify the allergen components of soybean and celery. The sensitivity test showed that the detection limit of soybean and celery components was 0.01. Therefore, this method can be used as a specific method for simultaneous detection of soybean and celery allergens.
