DEHP、MEHP對(duì)小鼠胚胎干細(xì)胞細(xì)胞毒性的研究

【摘要】:目的分析不同濃度的鄰苯二甲酸二乙基己酯[di(2-ethylhexyl)phthalate,DEHP]及其活性代謝產(chǎn)物鄰苯二甲酸單乙基己基酯[mono(2-ethylhexyl)phthalate,MEHP)對(duì)小鼠胚胎干細(xì)胞細(xì)胞毒性、增殖的影響。方法用含有不同濃度DEHP(1 000、100、10、1、0.1、0.01μmol/L)、MEHP(1 000、100、10、1、0.1、0.01μmol/L)的培養(yǎng)液,對(duì)胚胎干細(xì)胞進(jìn)行培養(yǎng),用CCK-8法檢測(cè)細(xì)胞活性。結(jié)果染毒96 h后,高劑量(1 000μmol/L)DEHP、MEHP均對(duì)胚胎干細(xì)胞的增殖有抑制作用(P0.05),且染毒劑量與細(xì)胞抑制率之間存在明顯的正相關(guān)性,相關(guān)系數(shù)分別為0.51、0.62(P0.05)。結(jié)論在一定濃度范圍內(nèi),DEHP及MEHP均對(duì)小鼠ESC具有細(xì)胞毒性,且有濃度依賴性。
[Abstract]:Objective to study the effects of di (2-ethylhexyl) phthalate,DEHP and its active metabolite, mono (2-ethylhexyl) phthalate,MEHP, on the toxicity and proliferation of mouse embryonic stem cells. Methods embryonic stem cells were cultured in culture medium containing different concentrations of DEHP (1 000 DEHP (0. 1 0. 01 渭 mol/L), MEHP (1 000) and 0. 1 0. 01 渭 mol/L). The activity of embryonic stem cells was detected by CCK-8 method. Results after 96 h exposure, high dose (1 000 渭 mol/L) DEHP,MEHP could inhibit the proliferation of embryonic stem cells (P0.05), and there was a significant positive correlation between the dose and the cell inhibition rate, the correlation coefficient was 0.51 渭 mol/L (P0.05). Conclusion in a certain concentration range, both DEHP and MEHP have cytotoxicity to ESC in a concentration dependent manner.
