微核、染色體試驗用乙醇固定淋巴細(xì)胞可行性研究

【摘要】:目的探討微核、染色體試驗中用乙醇部分代替甲醇固定淋巴細(xì)胞的可行性及相關(guān)技術(shù)方法。方法采用周圍血半微量全血常規(guī)培養(yǎng)法進(jìn)行試驗。試驗按固定液中甲醇和乙醇的比例不同分為Ⅰ、Ⅱ、Ⅲ、Ⅳ組,4種方法固定液中甲醇、乙醇和冰醋酸的比例分別為3∶0∶1、2∶1∶1、1∶2∶1、0∶3∶1(V∶V∶V),其他條件和操作均相同。取周圍靜脈血適量接種于RPMI-1640培養(yǎng)基中,于37℃培養(yǎng)72 h(染色體培養(yǎng)48 h),用體積分?jǐn)?shù)為10%吉姆薩染液染色。結(jié)果Ⅰ、Ⅱ和Ⅲ組淋巴細(xì)胞均分散良好、染色均勻適中,胞漿呈藍(lán)灰色,胞核呈紫紅色,胞核輪廓清楚、邊界清晰,均適合微核觀察計數(shù);Ⅳ組細(xì)胞容易聚集成團(tuán)、胞漿易于破裂,致使適合觀察的淋巴細(xì)胞數(shù)太少,從而影響微核的觀察計數(shù)。Ⅰ、Ⅱ和Ⅲ組之間淋巴細(xì)胞著色良好率比較,差異有統(tǒng)計學(xué)意義(P0.05);其中Ⅱ組淋巴細(xì)胞著色良好率高于Ⅲ組(P0.05)。Ⅰ、Ⅱ和Ⅲ組淋巴細(xì)胞胞漿完整率和淋巴細(xì)胞微核細(xì)胞率分別比較,差異均無統(tǒng)計學(xué)意義(P0.05)。Ⅰ、Ⅱ、Ⅲ和Ⅳ組淋巴細(xì)胞細(xì)胞成團(tuán)指數(shù)比較,差異有統(tǒng)計學(xué)意義(P0.05);其中Ⅰ、Ⅱ和Ⅲ組細(xì)胞成團(tuán)指數(shù)均低于Ⅳ組(P0.05)。Ⅰ、Ⅱ和Ⅲ組淋巴細(xì)胞染色體呈紫紅色、分散良好、大小適中、背景清楚,均適合染色體觀察;Ⅳ組不適用于染色體觀察。結(jié)論微核和染色體試驗的固定液中加入一定比例的乙醇(乙醇與甲醇的體積比不超過2∶1)代替甲醇固定淋巴細(xì)胞,不影響微核和染色體的觀察,并可降低甲醇中毒風(fēng)險。
