氯仿和鄰苯二甲酸二辛酯聯(lián)合染毒對小鼠肝臟酶的影響

【摘要】:目的觀察氯仿(chloroform,CF)和鄰苯二甲酸二辛酯(dioctylphthalate,DOP)聯(lián)合染毒對小鼠肝臟酶的影響。方法選取40只成年ICR小鼠,隨機分成8組,即雌性對照組(玉米油)、雌性DOP單獨染毒組(1/10 LD_(50),540 mg/kg)、雌性CF單獨染毒組(1/10 LD_(50),112 mg/kg)、雌性DOP(1/10 LD_(50),540 mg/kg)+CF(1/10 LD_(50),112 mg/kg)聯(lián)合染毒組,雄性也做同樣分組及處理,每組5只。采用灌胃方式連續(xù)染毒15 d,每天1次。測定肝臟組織中乳酸脫氫酶(LDH)、ATP酶活力。結(jié)果與對照組比較,DOP組、CF組和聯(lián)合染毒組小鼠肝臟器系數(shù)、LDH活力均升高,Na~+-K~+-ATP酶、Ca~(2+)-Mg~(2+)-ATP酶活力均降低,差異有統(tǒng)計學(xué)意義(P0.05)。雄性聯(lián)合染毒組肝臟器系數(shù)均高于DOP單獨染毒組、CF單獨染毒組,差異有統(tǒng)計學(xué)意義(P0.05);雌性聯(lián)合染毒組與DOP單獨染毒組的肝臟器系數(shù)無明顯差異(P0.05),高于CF單獨染毒組,差異有統(tǒng)計學(xué)意義(P0.05)。與DOP單獨染毒組比較,聯(lián)合染毒組雌性小鼠的LDH、Na~+-K~+-ATP酶活力均無明顯差異(P0.05),Ca~(2+)-Mg~(2+)-ATP酶活力升高,差異有統(tǒng)計學(xué)意義(P0.05);與CF單獨染毒組比較,聯(lián)合染毒組雌性小鼠LDH活力升高,Ca~(2+)-Mg~(2+)-ATP酶活力下降,差異均有統(tǒng)計學(xué)意義(P0.05),Na~+-K~+-ATP酶活力無明顯差異(P0.05)。與DOP單獨染毒組比較,聯(lián)合染毒組雄性小鼠的Ca~(2+)-Mg~(2+)-ATP、Na~+-K~+-ATP酶活力均無明顯差異(P0.05),LDH酶活力升高,差異有統(tǒng)計學(xué)意義(P0.05);與CF單獨染毒組比較,聯(lián)合染毒組雄性小鼠的LDH活力升高,Na~+-K~+-ATP酶、Ca~(2+)-Mg~(2+)-ATP酶活力下降,差異均有統(tǒng)計學(xué)意義(P0.05)。雄性聯(lián)合染毒對肝臟器系數(shù)的影響尚未表現(xiàn)出聯(lián)合作用(P0.05),其他各聯(lián)合組均存在聯(lián)合作用(P0.05),表現(xiàn)為拮抗作用。結(jié)論DOP和CF聯(lián)合染毒可能對小鼠肝臟系統(tǒng)的無氧糖酵解和能量代謝產(chǎn)生抑制作用。
[Abstract]:Objective to observe the effect of chloroform (chloroform,CF) combined with dioctyl phthalate (dioctylphthalate,DOP) on liver enzymes in mice. Methods Forty adult ICR mice were randomly divided into 8 groups: female control group (corn oil), female DOP alone (1 / 10 LD_ (50) 540 mg/kg), female CF alone (1 / 10 LD_ (50) 1 / 112 mg/kg) and female DOP (1 / 10 LD_ (50) 540mg/kg) CF (1 / 10 LD_ (50) 112mg/kg). The males were also divided into groups and treated with 5 rats in each group. The rats were administrated by gavage for 15 days, once a day. The activity of lactate dehydrogenase (LDH) ATPase in liver tissue was measured. Results compared with the control group, the liver organ coefficient and LDH activity in the DOP group and the combined exposure group were increased, and the activities of Na ~ (2) -mg ~ (2) -ATPase decreased significantly (P0.05). The liver organ coefficient of male combined exposure group was higher than that of DOP alone group (P0.05), and the liver organ coefficient of female combined exposure group and DOP alone group had no significant difference (P0.05), which was higher than that of CF alone group. The difference was statistically significant (P0.05). There was no significant difference in the activity of LDH,Na~ -K ~ -ATPase in female mice exposed to DOP alone (P0.05), the activity of Ca2 -Mg2 -ATPase increased significantly (P0.05), and the activity of LDH,Na~ -K ~ -ATPase was significantly higher than that of CF alone (P0.05), and compared with that of CF alone, the activity of LDH,Na~ -K ~ (2) -ATPase in the combined group was significantly higher than that in the control group (P0.05). The LDH activity of female mice in combined exposure group increased and the activity of Ca ~ (2) -mg ~ (2) -ATPase decreased, there was no significant difference (P0.05) in Na ~ -K ~ -ATPase activity (P0.05). There was no significant difference in Ca~ (2) -mg ~ (2) -ATPase activity of Na- K ~ -ATPase between the combined exposure group and the DOP alone group (P0.05), the difference was statistically significant (P0.05), and compared with the CF alone group, there was no significant difference in Ca~ _ (2) -mg ~ (2) -ATPase activity between the two groups. The LDH activity of the combined exposure group increased with the decrease of Ca ~ (2) -mg ~ (2) -ATPase activity of Na ~ -K ~ -ATPase (P0.05). The effect of male combined exposure on liver organ coefficient has not yet shown a combined effect (P0.05), other combined groups have combined effects (P0.05), showing antagonism. Conclusion combined exposure of DOP and CF may inhibit anaerobic glycolysis and energy metabolism in mouse liver system.
