砷對HOXD10基因表達(dá)的影響

【摘要】:目的探討砷對周圍血淋巴細(xì)胞中及人肺腺癌細(xì)胞系A(chǔ)549細(xì)胞中同源異型盒D10(HOXD10)基因表達(dá)的影響。方法 (1)采用判斷抽樣方法,抽取某砒霜廠職業(yè)性接觸砷的59名工人為接觸組,以無職業(yè)性接觸砷的17名當(dāng)?shù)鼐用駷閷φ战M。采用氫化物發(fā)生-冷肼捕集-原子吸收法檢測2組人群尿中形態(tài)砷水平,采用實時熒光定量聚合酶鏈?zhǔn)椒磻?yīng)(qRT-PCR)法檢測周圍血淋巴細(xì)胞中HOXD10的表達(dá)。(2)以濃度為0.0、0.1、0.5、1.0、2.0μmol/L的三氧化二砷(As_2O_3)處理A549細(xì)胞,采用比色法檢測細(xì)胞存活率,采用qRT-PCR法檢測HOXD10的表達(dá)。結(jié)果 (1)接觸組人群尿中無機(jī)砷、一甲基砷酸、二甲基砷酸、總砷水平和周圍血淋巴細(xì)胞中HOXD10 mRNA相對表達(dá)水平均高于對照組(P0.05)。(2)As_2O_3可導(dǎo)致A549細(xì)胞存活率呈現(xiàn)劑量依賴性下降(P0.01),HOXD10mRNA相對表達(dá)水平呈現(xiàn)劑量依賴性上調(diào)(P0.01)。A549細(xì)胞存活率和HOXD10 mRNA相對表達(dá)水平呈負(fù)相關(guān),相關(guān)系數(shù)為-0.777(P0.01)。結(jié)論砷可導(dǎo)致職業(yè)接觸人群周圍血中HOXD10表達(dá)上調(diào)。As_2O_3可抑制A549細(xì)胞增殖,可能與上調(diào)HOXD10表達(dá)有關(guān)。

(2) A549 cells were treated with arsenic trioxide (As_2O_3) at a concentration of 0. 01 渭 mol/L. The survival rate of A549 cells was detected by colorimetric assay and the expression of HOXD10 was detected by qRT-PCR assay. Results (1) Inorganic arsenic, monomethyl arsenic acid, dimethyl arsenic acid in urine of exposed group, The total arsenic level and the relative expression of HOXD10 mRNA in peripheral blood lymphocytes were higher than those in the control group (P0.05). (2) As_2O_3 could lead to a dose-dependent decrease in the survival rate of A549 cells (P0.01). The relative expression level of HOXD10 mRNA in A549 cells was up-regulated in a dose-dependent manner (P0.01). A549 cell survival rate and HOXD10 were increased in a dose-dependent manner. The relative expression level of mRNA was negatively correlated. The correlation coefficient was -0.777 (P0.01). Conclusion arsenic can induce the upregulation of HOXD10 expression in blood of occupational exposed population. As _ 2O _ 3 can inhibit the proliferation of A549 cells, which may be related to the up-regulation of HOXD10 expression.
