【摘要】：目的研究α、β、γ、δ-生育酚對四氯化碳(CCl_4)肝損傷小鼠的保護(hù)作用。方法將60只健康雄性ICR小鼠隨機分成6組:正常組(NG)、CCl_4模型組(MG)、α-生育酚組(α-T)、β-生育酚組(β-T)、γ-生育酚組(γ-T)和δ-生育酚組(δ-T)。各生育酚組分別給予相應(yīng)的羧甲基纖維素鈉(CMC-Na)生育酚混懸液[100 mg/(kg·d)],NG組和MG組每日給予等量CMC-Na溶液灌胃,連續(xù)2w。末次灌胃2h后,正常組腹腔注射橄欖油,其余各組腹腔注射0.5%CCl_4橄欖油溶液。16h后處死小鼠,收集血清和肝臟。試劑盒測定小鼠血清谷丙轉(zhuǎn)氨酶(ALT)、總膽固醇(TC)、甘油三酯(TG)水平及肝組織丙二醛(MDA)、超氧化物歧化酶(SOD)、谷胱甘肽過氧化物酶(GSH-Px)、過氧化氫酶(CAT)的含量;熒光定量法測定肝臟SOD、CAT、GSH-Px基因相對表達(dá)量;光學(xué)顯微鏡觀察小鼠肝臟病理學(xué)變化。結(jié)果與MG組相比,α-T組小鼠血清ALT、TG水平及肝臟MDA含量均顯著降低,肝臟SOD、GSH-Px、CAT活力及m RNA表達(dá)量均顯著升高;β-T組小鼠血清ALT、TG水平及肝臟MDA水平均顯著降低,肝臟SOD水平顯著升高;γ、δ-T組小鼠肝臟MDA水平亦顯著降低。小鼠肝臟病理切片觀察顯示,各生育酚組較MG組有不同程度的改善。結(jié)論生育酚異構(gòu)體對四氯化碳肝損傷小鼠有不同程度的保護(hù)作用,α-生育酚和β-生育酚的效果較好,其機理可能與其抗氧化作用相關(guān)。
[Abstract]:Objective to study the protective effect of 偽, 尾, 緯, 未 -tocopherol on carbon tetrachloride (CCl_4) liver injury in mice. Methods 60 healthy male ICR mice were randomly divided into 6 groups: normal group, (NG) group, (MG), 偽 -tocopherol group (偽 -T), 尾 -tocopherol group (尾 -T), 緯 -tocopherol group (緯 -T) and 未 -tocopherol group (未 -T). Each tocopherol group was given corresponding carboxymethyl cellulose sodium (CMC-Na) tocopherol suspension [100 mg/ (kg d)] NG group and MG group by intragastric administration of the same amount of CMC-Na solution daily for 2 weeks. Two hours after the last gastric administration, the normal group was injected with olive oil intraperitoneally, the other groups were injected with 0.5l_4 olive oil solution for 16 h, then the mice were killed and the serum and liver were collected. The levels of serum alanine aminotransferase (ALT), total cholesterol (TC), triglyceride (TG), malondialdehyde (MDA) (MDA), superoxide dismutase (MDA), (SOD), glutathione peroxidase (GSH-Px) and catalase (CAT) (CAT) were measured. The relative expression of SOD,CAT,GSH-Px gene in liver was measured by fluorescence quantitative method, and the pathological changes of mouse liver were observed by optical microscope. Results compared with MG group, serum ALT,TG level and liver MDA content in 偽 -T group were significantly decreased, liver SOD,GSH-Px,CAT activity and m RNA expression were significantly increased, serum ALT,TG level and liver MDA level were significantly decreased in 尾 -T group. The level of SOD in liver increased significantly, and the level of MDA in liver of 緯, 未-T group decreased significantly. The pathological sections of mouse liver showed that each tocopherol group was better than MG group to some extent. Conclusion tocopherol isomers have different protective effects on carbon tetrachloride liver injury in mice. 偽 -tocopherol and 尾 -tocopherol have better effects, and the mechanism may be related to their antioxidant effects.
