【摘要】：目的研究1,1-二氨基-2,2-二硝基乙烯(FOX-7)對(duì)大鼠的亞慢性經(jīng)口毒性效應(yīng)。方法無(wú)特定病原體級(jí)健康成年SD大鼠隨機(jī)分為對(duì)照組和低、中、高劑量組,每組24只,雌雄各半。3個(gè)劑量組分別以10、30和90 mg/(kg·d)體質(zhì)量FOX-7-水淀粉(水淀粉質(zhì)量分?jǐn)?shù)為4.0%)混懸液灌胃染毒,1次/d,每周6 d,連續(xù)90 d;對(duì)照組給予等體積水淀粉溶液灌胃。觀察大鼠的中毒表現(xiàn)、體質(zhì)量、食物利用率、血常規(guī)、血液生化指標(biāo)、臟器系數(shù)變化及組織器官的組織病理學(xué)改變。結(jié)果 1雌性、雄性高劑量組大鼠在染毒第28天后體質(zhì)量均低于同時(shí)間點(diǎn)同性別對(duì)照組(P0.05),在染毒第77和90天食物利用率分別低于同時(shí)間點(diǎn)同性別對(duì)照組(P0.05)。2與雌性對(duì)照組比較,雌性低、中和高劑量組大鼠紅細(xì)胞計(jì)數(shù)、血紅蛋白、紅細(xì)胞壓積均下降(P0.05),雌性高劑量組大鼠血小板計(jì)數(shù)下降(P0.05)。與雄性對(duì)照組比較,雄性高劑量組大鼠紅細(xì)胞計(jì)數(shù)、血紅蛋白、紅細(xì)胞壓積和平均紅細(xì)胞血紅蛋白濃度均下降(P0.05),雄性中和高劑量組大鼠血小板計(jì)數(shù)均下降(P0.05)。3與雌性對(duì)照組比較,雌性中和高劑量組大鼠血清總膽固醇水平均升高(P0.05),雌性高劑量組大鼠尿素氮水平升高(P0.05)。與對(duì)照組比較,高劑量組大鼠總蛋白和尿酸水平均升高,而乳酸脫氫酶活力下降(P0.05)。4雌性高劑量組大鼠脾臟臟器系數(shù)高于雌性對(duì)照組(P0.05),雄性中和高劑量組大鼠脾臟臟器系數(shù)均高于雄性對(duì)照組(P0.05);高劑量組大鼠肝臟和腎臟的臟器系數(shù)均高于對(duì)照組(P0.05);雄性高劑量組大鼠睪丸、附睪的臟器系數(shù)均低于雄性對(duì)照組(P0.05),且均出現(xiàn)不同程度的睪丸曲細(xì)精管萎縮,精細(xì)胞減少。5未觀察到有害作用水平在雌性大鼠為小于10.00 mg/(kg·d),雄性大鼠為10.00 mg/(kg·d)。結(jié)論在本實(shí)驗(yàn)條件下,FOX-7可抑制大鼠的生長(zhǎng)發(fā)育,引起血液系統(tǒng)及雄鼠的生殖系統(tǒng)損害。
[Abstract]:Objective to study the subchronic oral toxicity of 1-diamino-2-dinitroethylene-2-dinitroethylene-7 (FOX-7) in rats. Methods healthy adult SD rats without specific pathogens were randomly divided into control group and low, medium and high dose groups with 24 rats in each group. Three dose groups were treated with FOX-7- water starch (water starch 4.0wt%) respectively, 6 days a week for 90 days, and the control group was given equal volume water starch solution for 90 days. The toxic manifestation, body mass, food utilization rate, blood routine, blood biochemical index, organ coefficient and histopathological changes were observed. Results 1 the body mass of female and male rats in the high dose group was lower than that of the control group at the same time after 28 days of exposure, and the food availability on the 77th and 90th day of exposure was lower than that of the female control group at the same time. The red blood cell count, hemoglobin and hematocrit of female rats in low, middle and high dose groups all decreased P0.05A, and the platelet count of female high-dose rats decreased P0.05a. Compared with the male control group, the red blood cell count, hemoglobin, hematocrit and mean erythrocyte hemoglobin concentration in the male high dose group all decreased P0.05, and the platelet count in the male middle and high dose group were all decreased compared with the female control group. The serum total cholesterol levels of female middle and high dose groups were all higher than that of female high dose group, and the level of urea nitrogen in female high dose group was higher than that of female high dose group. Compared with the control group, the levels of total protein and uric acid in the high dose group were higher than those in the control group. The spleen organ coefficient of female rats in high dose group was higher than that of female control group, the spleen organ coefficient of male middle and high dose group was higher than that of male control group, and that of liver and kidney of high dose group was higher than that of male control group. The visceral coefficients were higher than those of the control group (P0.05A), and the testis of the male rats in the high dose group, The organ coefficient of epididymis was lower than that of male control group (P 0.05), and testicular seminiferous tubule atrophy was observed to some extent, and no harmful effects were observed in female rats and male rats. Conclusion FOX-7 can inhibit the growth and development of rats and cause damage to the blood system and reproductive system of male rats.
