【摘要】：目的觀察鎘染毒對去卵巢大鼠腎臟中雌激素受體α(ERα)、雌激素受體β(ERβ)表達的影響。方法將24只健康12周齡SPF級SD雌性大鼠隨機分為3組,分別為假手術(shù)(蒸餾水)組、去卵巢(蒸餾水)組、染鎘(200 mg/L)去卵巢組,每組8只。假手術(shù)組切除小塊卵巢周圍的脂肪,去卵巢組和染鎘去卵巢組切除雙側(cè)卵巢。術(shù)后1周,采用自由飲水方式進行染毒,連續(xù)染毒12周。采用電感耦合等離子體質(zhì)譜法測定大鼠血鎘、尿鎘水平,采用酶聯(lián)免疫吸附測定血清雌二醇、β2-微球蛋白水平,采用Western blot技術(shù)檢測腎臟組織中ERα、ERβ蛋白的表達水平。結(jié)果與假手術(shù)組相比,去卵巢組染鎘組12周末體重下降(P0.05)、腎臟系數(shù)升高(P0.05),去卵巢組及染鎘去卵巢組雌激素水平降低(P0.05);與去卵巢組相比,染鎘去卵巢組體重、雌激素水平降低(P0.05),腎臟系數(shù)、β2-微球蛋白、尿鎘及血鎘水平升高(P0.05);染鎘去卵巢組血鎘與尿鎘、β2-微球蛋白、腎臟ERβ的表達水平正相關(guān)(P0.05),而與ERα無統(tǒng)計學(xué)相關(guān)性(P0.05)。結(jié)論重金屬鎘可降低去卵巢大鼠血清雌激素水平及增加雌激素受體β受體表達,這可能是鎘的腎毒性作用機制之一。
[Abstract]:Objective to observe the effect of cadmium exposure on the expression of estrogen receptor 偽 -ER 偽 and estrogen receptor 尾 -ER 尾 in the kidney of ovariectomized rats. Methods Twenty-four healthy 12-week-old SPF female rats were randomly divided into three groups: sham-operated (distilled water) group, ovariectomized (distilled water) group and ovariectomized group (200mg / L). In sham-operated group, fat around small ovary was removed, ovariectomized group and cadmium stained ovariectomized group. One week after operation, free drinking water was used for 12 weeks. Serum cadmium and urine cadmium levels were measured by inductively coupled plasma mass spectrometry (ICP-MS), serum estradiol and 尾 _ 2-microglobulin were measured by enzyme-linked immunosorbent assay (Elisa), and ER 偽 -ER 尾 protein expression in renal tissue was detected by Western blot technique. Results compared with sham-operation group, the weight of ovariectomized group decreased P0.05, the kidney coefficient increased P0.05, the estrogen level of ovariectomized group and cadmium-exposed ovariectomized group decreased P0.05, compared with ovariectomized group, the body weight of cadmium-exposed ovariectomized group was lower than that of ovariectomized group. Estrogen level decreased P0.05G, renal coefficient, 尾 2-microglobulin, urine cadmium and blood cadmium level increased P0.050.The expression level of blood cadmium and urine cadmium, 尾 2-microglobulin and renal ER 尾 was positively correlated with the expression level of ER 尾 in ovariectomized group (P 0.05), but not correlated with ER 偽 (P 0.05). Conclusion cadmium can decrease the serum estrogen level and increase the expression of estrogen receptor 尾 receptor in ovariectomized rats, which may be one of the mechanisms of renal toxicity of cadmium.
