【摘要】：目的觀察環(huán)磷酸腺苷(cAMP)信號(hào)在大鼠矽肺纖維化發(fā)生、發(fā)展過程中的變化及其對(duì)矽肺纖維化形成的影響。方法采用HOPE-MED8050動(dòng)式染塵控制系統(tǒng)復(fù)制大鼠矽肺模型,雄性Wistar大鼠隨機(jī)分為6組,每組10只,分別染塵0、2、4、8、12和16周。采用Masson三色染色觀察肺組織形態(tài),采用免疫組織化學(xué)法染色和Western blot檢測(cè)α-平滑肌肌動(dòng)蛋白(α-SMA)、Ⅰ型膠原(CollagenⅠ)、纖連蛋白(Fn)、激動(dòng)型G蛋白α(Gαs)、抑制型G蛋白α(Gαi2、Gαi3)及cA MP的表達(dá)。結(jié)果 Masson三色染色顯示,矽肺模型4周組細(xì)胞性結(jié)節(jié)區(qū)域可見藍(lán)紫色膠原沉積,并隨模型制備時(shí)間的延長纖維化病變區(qū)域膠原沉積逐漸增加。免疫組織化學(xué)法染色顯示,對(duì)照組α-SMA陽性表達(dá)部位在氣管及血管平滑肌;在矽肺模型16周組,α-SMA表達(dá)于結(jié)節(jié)周邊及間質(zhì)纖維化病變區(qū)域。α-SMA、CollagenⅠ、Fn及Gαi2、Gαi3蛋白表達(dá)在矽肺模型4、8、12和16周較對(duì)照組逐漸增多。在矽肺模型8周組Gαs蛋白表達(dá)及cAMP含量較對(duì)照組下降,至染塵16周達(dá)最低。結(jié)論 cAMP的表達(dá)在矽肺發(fā)生、發(fā)展過程中呈下降趨勢(shì),cAMP信號(hào)參與大鼠矽肺纖維化過程。
[Abstract]:Aim to observe the changes of cyclic adenosine monophosphate (camp) signal in the pathogenesis and development of silicosis in rats and its effect on the formation of silicosis. Methods the silicosis model of rats was induced by HOPE-MED8050 dynamic dust control system. Male Wistar rats were randomly divided into 6 groups with 10 rats in each group for 12 and 16 weeks respectively. The expression of 偽 -smooth muscle actin (偽 -SMAA), fibronectin (FN), activated G protein 偽 G 偽 sn, inhibitory G protein 偽 G 偽 I 2G 偽 I 3 and camp were detected by immunohistochemical staining and Western blot staining. Results Masson trichrome staining showed that blue-purple collagen deposition could be seen in the cellular nodules of the silicosis model for 4 weeks, and the collagen deposition increased gradually with the prolongation of the time of the establishment of the model. Immunohistochemical staining showed that the positive expression of 偽 -SMA in the control group was located in trachea and vascular smooth muscle.
