丙烯酰胺暴露對幼鼠胼胝體PLP和MBP表達的影響

【摘要】:目的:檢測斷乳期幼鼠胼胝體髓鞘蛋白PLP和MBP的表達,探討丙烯酰胺(acrylamide,ACR)染毒對幼鼠胼胝體部髓鞘發(fā)育的影響。方法:斷乳期幼鼠隨機分為對照組(0 mg/kg)、低(18 mg/kg)和高(36 mg/kg)劑量組,每組12只,從出生后第22~42 d進行灌胃染毒。觀測幼鼠步態(tài)的變化,用免疫組化方法和免疫熒光雙標(biāo)記法檢測幼鼠胼胝體髓鞘蛋白脂蛋白(myelin PLP,PLP)和髓鞘堿性蛋白(myelin basic protein,MBP)的表達。結(jié)果:ACR染毒后幼鼠的步態(tài)評分均增加,差異有統(tǒng)計學(xué)意義(P0.01),免疫組織化學(xué)檢測結(jié)果顯示,與對照組相比較,ACR高劑量組幼鼠大腦PLP和MBP表達減少,差異有統(tǒng)計學(xué)意義(P0.05)。免疫熒光雙標(biāo)技術(shù)檢測結(jié)果與免疫組織化學(xué)檢測結(jié)果一致,即ACR染毒后胼胝體PLP和MBP均表達減少。結(jié)論:ACR染毒可能會通過減少PLP和MBP的表達,抑制胼胝體髓鞘的形成而影響神經(jīng)系統(tǒng)發(fā)育。
[Abstract]:Aim: to detect the expression of myelin PLP and MBP in corpus callosum of young rats during weaning period and to investigate the effect of acrylamide acrylamide on the development of myelin in corpus callosum of young rats.Methods: young rats during weaning period were randomly divided into control group (0 mg / kg), low dose (18 mg / kg) and high dose group (36 mg / kg). 12 rats in each group were given intragastric administration from 22 to 42 days after birth.The changes of gait of young rats were observed, and the expression of myelin basic (PLP) and myelin basic protein (MBP) in corpus callosum protein (PLP) and myelin basic protein (myelin basic) in young rats were detected by immunohistochemistry and immunofluorescence double labeling method.Results the gait scores of the young rats were all increased after the treatment with 1: ACR, and the difference was statistically significant (P 0.01). The results of immunohistochemistry showed that the expression of PLP and MBP in the brain of the high dose group was lower than that of the control group, and the difference was statistically significant (P 0.05).The results of double immunofluorescence staining were consistent with those of immunohistochemistry, that is, the expression of PLP and MBP in corpus callosum decreased after ACR exposure.ConclusionACR exposure may affect the development of nervous system by reducing the expression of PLP and MBP and inhibiting the formation of corpus callosum myelin.
