鄰苯二甲酸單乙基己基酯對(duì)原代培養(yǎng)新生雌性大鼠下丘腦神經(jīng)元生殖神經(jīng)內(nèi)分泌相關(guān)基因表達(dá)影響

【摘要】:目的探討鄰苯二甲酸單乙基己基酯(mono-2-ethylhexyl phthalate,MEHP)對(duì)新生雌性大鼠下丘腦生殖神經(jīng)內(nèi)分泌功能的影響。方法對(duì)出生24h內(nèi)的新生雌性SD大鼠下丘腦神經(jīng)元進(jìn)行原代培養(yǎng),分別暴露于含終濃度為0(對(duì)照)、10~(-15)、10~(-12)、10~(-9)、10~(-6)mol/L MEHP的培養(yǎng)基染毒24h。采用qRT-PCR法檢測(cè)Gn RH、Gn RHr、Kiss1及Kiss1r基因的表達(dá)水平。結(jié)果與對(duì)照組相比,各濃度MEHP暴露組原代培養(yǎng)新生雌性大鼠下丘腦神經(jīng)元Kiss1基因的表達(dá)水平均較低,差異有統(tǒng)計(jì)學(xué)意義(P0.05);而Gn RH、Gn RHr和Kiss1r基因的表達(dá)水平均無明顯改變。結(jié)論 MEHP有可能通過下調(diào)新生雌性大鼠下丘腦神經(jīng)元Kiss1基因表達(dá)水平來干擾生殖神經(jīng)的內(nèi)分泌調(diào)控功能。
[Abstract]:Objective to investigate the effect of monoethyl hexyl phthalate mono-2-ethylhexyl phthalate (MEHP) on the reproductive neuroendocrine function of hypothalamus in neonatal female rats. Methods the hypothalamic neurons of newborn female SD rats within 24 hours were cultured in primary culture. The expression levels of Gn RHN Gn RHrN Kiss1 and Kiss1r gene were detected by qRT-PCR assay. The expression level of Kiss1 gene in hypothalamic neurons of primary cultured neonatal female rats was lower in MEHP exposure group. The difference was statistically significant (P 0.05), but the expression levels of Gn RHN RHr and Kiss1r genes were not significantly changed. Conclusion MEHP may interfere with the endocrine regulation of reproductive nerve by down-regulating the expression level of Kiss1 gene in hypothalamic neurons of newborn female rats.
