16S rDNA克隆文庫法分析兩種魷魚墨多糖對小鼠腸道菌群多樣性的影響

【摘要】:探討北太平洋魷魚墨多糖與秘魯魷魚墨多糖對化療模型小鼠腸道菌群的影響。將40只Balb/c小鼠隨機分為4組:正常組、模型組、北太平洋魷魚墨多糖組、秘魯魷魚墨多糖組。采用環(huán)磷酰胺化療模型,無菌收集小鼠糞便并提取細(xì)菌基因組DNA,通過構(gòu)建各組小鼠腸道細(xì)菌的16S rDNA克隆文庫,單酶切分型,觀察魷魚墨多糖對腸道菌群的作用。試驗結(jié)果表明:兩種魷魚墨多糖對化療模型小鼠腸道菌群種類有一定調(diào)節(jié)作用,且兩種魷魚墨多糖效果不盡相同。
[Abstract]:To investigate the effects of the ink polysaccharides of squid from North Pacific and Peru on the intestinal flora of chemotherapeutic model mice, 40 Balb/c mice were randomly divided into four groups: normal group, model group, polysaccharide group of squid ink from North Pacific, and so on. Using cyclophosphamide (cyclophosphamide) chemotherapeutic model, the feces of mice were collected aseptically and the genomic DNA of bacteria was extracted. The 16s rDNA clone library of intestinal bacteria in each group was constructed and digested by single enzyme. The effects of squid ink polysaccharides on intestinal flora were observed. The results showed that the two kinds of squid ink polysaccharides could regulate the intestinal flora of chemotherapeutic model mice, and the effects of two kinds of squid ink polysaccharides were different.
