食蟹猴腹腔注射低劑量百草枯致胰腺α-突觸核蛋白的病理變化

【摘要】:本研究旨在通過食蟹猴經(jīng)腹腔注射低劑量百草枯來探討胰腺α-突觸核蛋白(α-synuclein,α-syn)的病理變化。由廣西南寧靈康賽諾科生物科技有限公司提供的12只(8~10歲)健康雄性食蟹猴為實(shí)驗(yàn)動(dòng)物,隨機(jī)分為實(shí)驗(yàn)組A、B和正常對(duì)照組C,每組4只,實(shí)驗(yàn)組分別于給藥百草枯(paraquat,PQ)5個(gè)月,2個(gè)月后安樂死(對(duì)照組每階段2只)。利用免疫組織化學(xué)方法和免疫蛋白印記方法檢測(cè)各組食蟹猴胰島α-syn的表達(dá)特點(diǎn)和表達(dá)量。結(jié)果顯示,α-syn免疫活性物質(zhì)主要見于胰島細(xì)胞胞漿,并且α-syn在各組食蟹猴胰島存在差異表達(dá),其中給藥百草枯5個(gè)月后的食蟹猴胰島α-syn表達(dá)量最高,對(duì)照組表達(dá)量最低。我們的研究表明食蟹猴腹腔注射低劑量百草枯可致胰島α-syn表達(dá)上調(diào),百草枯是造成胰島腺α-syn差異表達(dá)的關(guān)鍵因素之一,其結(jié)論將為相關(guān)疾病的模型制作、早期診斷及干預(yù)提供新的思路。
[Abstract]:The aim of this study was to investigate the pathological changes of 偽 -synuclein (偽 -synn) in pancreas by intraperitoneal injection of paraquat into the abdominal cavity of crab-eating monkeys. 12 healthy male food samples were provided by Guangxi Nanning Ling Kangsanoki Biotechnology Co., Ltd. The crab monkey is an experimental animal. The rats were randomly divided into two groups: experimental group (n = 4) and control group (n = 4). The experimental group was treated with paraquat PQ for 5 months. After 2 months of euthanasia (control group, 2 rats in each stage), the expression of 偽 -syn in pancreatic islets of each group was detected by immunohistochemical method and immunoglobulin imprinting method. The results showed that 偽 -syn immunoreactive substances were mainly found in the cytoplasm of islet cells. The expression of 偽 -syn in the islets of crab-eating monkeys was the highest after 5 months of administration of paraquat, and the expression of 偽 -syn in the islets of crab-eating monkeys was the highest. Our study showed that the expression of 偽 -syn in pancreatic islets was up-regulated by intraperitoneal injection of paraquat, and paraquat was one of the key factors causing the differential expression of 偽 -syn in pancreatic islets. Early diagnosis and intervention provide new ideas.
