金納米—汞復(fù)合物檢測(cè)汞和金屬硫蛋白的新方法研究

【摘要】:汞是一種嚴(yán)重影響人類(lèi)健康的重金屬,已被多個(gè)國(guó)家列為優(yōu)先控制的環(huán)境污染物。汞現(xiàn)有的檢測(cè)方法大多操作繁瑣、儀器昂貴,因此,建立靈敏、簡(jiǎn)單、選擇性高的新方法,對(duì)于環(huán)境中汞污染的監(jiān)測(cè)和安全評(píng)價(jià)等均有重要意義。 本文第二章建立了基于金納米和四甲基聯(lián)苯胺(TMB)催化比色檢測(cè)汞離子的方法。在pH7.0的TAE緩沖液中,金納米粒子與汞離子反應(yīng)生成AuNPs-Hg過(guò)氧化物模擬酶,催化TMB顯色,在9.14×10-8~7.69×10-6mol·L-1范圍內(nèi),ΔA與汞離子濃度呈良好線性關(guān)系,回歸方程為ΔA=0.001+0.091c(×10-6mol·L-1),,相關(guān)系數(shù)為0.996,檢出限為2.74×10-8mol·L-1。該方法簡(jiǎn)便、快速、穩(wěn)定性好。 本文第三章建立了基于金納米-汞-TMB催化比色檢測(cè)金屬硫蛋白的新方法。在pH7.0的TAE緩沖液中,金屬硫蛋白和金納米粒子競(jìng)爭(zhēng)結(jié)合汞離子,抑制AuNPs-Hg過(guò)氧化物模擬酶的生成,減弱TMB催化反應(yīng),在5.93×10-9~9.23×10-8mol·L-1范圍內(nèi),ΔA與金屬硫蛋白濃度呈良好線性關(guān)系,回歸方程為ΔA=0.042+0.066c(×10-8mol·L-1),相關(guān)系數(shù)為0.990,檢出限為1.78×10-9mol·L-1。該方法簡(jiǎn)便、快速、靈敏、穩(wěn)定性好。 本文第四章建立了基于金納米和金屬硫蛋白紫外分光光度法檢測(cè)汞離子的新方法。在pH7.0的TAE緩沖液中,汞離子導(dǎo)致金納米-金屬硫蛋白復(fù)合物聚集,使得紫外吸收發(fā)生改變,且在5.6×10-8mol·L-1~2.0×10-6mol·L-1范圍內(nèi),體系(A626/A524)與汞離子的濃度有良好的線性關(guān)系,其回歸方程為(A626/A524)=-0.0132+0.4072c(×10-6mol·L-1),r=0.9961,檢出限為1.68×10-8mol·L-1。該方法簡(jiǎn)單、快速、靈敏。
[Abstract]:Mercury is a serious human health effects of heavy metals, many countries have been listed as priority pollutants. Most of the existing detection method of mercury complex operation, expensive equipment, therefore, to establish a sensitive, simple and new method for high selectivity, the significance of mercury pollution in the environment monitoring and safety evaluation and so on.
The second chapter of this paper established gold nanoparticles and four methyl benzidine (TMB) method based on catalytic colorimetric detection of mercury ions. In the TAE pH7.0 buffer, gold nanoparticles and mercury ions generated AuNPs-Hg mimetic peroxidase, catalytic TMB color, 9.14 x 10-8~7.69 x 10-6mol - L-1 range, there is a good linear the relationship between A and the concentration of mercury, the regression equation was A=0.001+0.091c (10-6mol * L-1), the correlation coefficient was 0.996, the detection limit of 2.74 * 10-8mol L-1. the method is simple, rapid, good stability.
The third chapter of this paper established a new method for colorimetric detection of metallothionein - Mercury gold nanoparticles catalyzed by -TMB. Based on TAE buffer pH7.0, metallothionein and gold nanoparticles binding mercury ion, simulating AuNPs-Hg peroxide enzyme inhibition, decreased TMB catalytic reaction in 5.93 * 10-9~9.23 * 10-8mol - L-1 range A, which showed a good linear relationship with metallothionein concentration, the regression equation was A=0.042+0.066c (10-8mol * L-1), the correlation coefficient was 0.990, the detection limit of 1.78 * 10-9mol L-1. the method is simple, rapid, sensitive and good stability.
A new method was established in the fourth chapter, gold nanoparticles and metallothionein based on UV spectrophotometric detection of mercury ion. In the TAE pH7.0 buffer, mercury ions lead to gold nano - metal sulfur protein complex aggregation, makes the ultraviolet absorption change, and in the 5.6 * 10-8mol L-1~2.0 * 10-6mol L-1 within the scope of the system (A626/A524) has a good linear relationship with the concentration of mercury ion, the regression equation was (A626/A524) =-0.0132+0.4072c (10-6mol * L-1), r=0.9961, a detection limit of 1.68 * 10-8mol L-1. the method is simple, rapid and sensitive.
