高效液相色譜法測定人血清中拉莫三嗪濃度的不確定度評定

【摘要】:目的對高效液相色譜(HPLC)法測定人血清中拉莫三嗪濃度的不確定度進(jìn)行評定。方法用HPLC法測定人血清中拉莫三嗪濃度,分析稱量、溶液配制、血清提取、標(biāo)準(zhǔn)曲線擬合及測量重復(fù)性等因素對結(jié)果的影響,計(jì)算各因素的不確定度及合成不確定度,并評定擴(kuò)展不確定度。結(jié)果 HPLC法測定人血清中拉莫三嗪低(1.50μg·mL~(-1))、中(8.00μg·mL~(-1))、高(20.00μg·mL~(-1))3個(gè)質(zhì)量濃度質(zhì)控樣品的擴(kuò)展不確定度分別為UL=0.30,UM=0.53,UH=0.98(P=95%,k=2);其相應(yīng)檢測濃度結(jié)果表示為(1.51±0.30),(8.22±0.53),(20.40±0.98)μg·mL~(-1)。結(jié)論本方法的相對測定不確定度主要是由重復(fù)性、線性擬合及提取回收過程引入,影響低濃度測定結(jié)果的因素主要是線性擬合及測量重復(fù)性。
[Abstract]:Objective to evaluate the uncertainty of determination of lamotriazine in human serum by high performance liquid chromatography (HPLC). Methods the HPLC method was used to determine the concentration of lamotriazine in human serum. The effects of weighing, solution preparation, serum extraction, standard curve fitting and repeatability on the results were analyzed, and the uncertainty of each factor and the synthetic uncertainty were calculated. The extended uncertainty is evaluated. Results the extended uncertainty for the determination of lamotriazine in human serum (1.50 渭 g 路mL ~ (- 1), (8.00 渭 g 路mL ~ (- 1),) (20.00 渭 g 路mL ~ (- 1) by HPLC was UL=0.30,. UM=0.53,UH=0.98 (P = 95%, k ~ (2); The corresponding detection concentration was (1.51 鹵0.30), (8.22 鹵0.53), (20.40 鹵0.98) 渭 g 路mL ~ (- 1). Conclusion the relative determination uncertainty of this method is mainly introduced by repeatability, linear fitting and extraction and recovery process. The main factors affecting the results of low concentration determination are linear fitting and measurement repeatability.
