【摘要】：目的:建立間苯三酚血藥濃度的測(cè)定方法,并研究間苯三酚顆粒在家兔體內(nèi)的藥動(dòng)學(xué)特征。方法:取6只健康家兔ig給予間苯三酚顆粒(80 mg/kg)溶液,于給藥前和給藥后0.083、0.17、0.25、0.3、0.33、0.42、0.5、0.75、1、1.5、2、3、4、6 h分別自耳動(dòng)脈采血1.5 ml。采用高效液相色譜法檢測(cè)其血藥濃度,色譜柱為Welchrom C18,流動(dòng)相為乙腈-磷酸鹽緩沖液(3.5∶96.5),流速為1.0 ml/min,柱溫為30℃,檢測(cè)波長(zhǎng)為267 nm,進(jìn)樣量為20μl,內(nèi)標(biāo)為對(duì)苯二酚。采用DAS 2.1.1藥動(dòng)學(xué)軟件計(jì)算其藥動(dòng)學(xué)參數(shù)。結(jié)果:間苯三酚在0.12~3.0μg/ml質(zhì)量濃度范圍內(nèi)與其峰面積與內(nèi)標(biāo)峰面積的比值呈良好的線性關(guān)系(r=0.999 3),提取回收率為86.02%~91.33%,RSD為4.9%~5.3%(n=3),日內(nèi)RSD為4.5%~5.6%(n=3),日間RSD為4.9%~5.8%(n=3);間苯三酚在家兔體內(nèi)的藥動(dòng)學(xué)參數(shù)t1/2β、tmax、cmax、AUC0-6 h分別為(1.05±0.16)h、(0.25±0.05)h、(1 076.01±19.62)μg/L、(919.02±34.00)μg·h/L。結(jié)論:該法專(zhuān)屬性強(qiáng)、精密度好,符合血漿樣品測(cè)定要求,可用于間苯三酚家兔體內(nèi)的藥動(dòng)學(xué)研究。間苯三酚顆粒在家兔體內(nèi)代謝過(guò)程符合一室模型。
