NT-3對哇巴因損傷耳蝸螺旋神經(jīng)節(jié)細胞后的影響

【摘要】:目的: 建立畦巴因選擇性損傷蒙古沙鼠耳蝸螺旋神經(jīng)節(jié)細胞的動物模型,在此基礎(chǔ)上研究耳蝸螺旋神經(jīng)節(jié)細胞損傷后,神經(jīng)營養(yǎng)因子-3對耳蝸螺旋神經(jīng)節(jié)細胞的保護作用,并研究探討其神經(jīng)保護作用機理。 方法: 將聽力正常的雄性蒙古沙鼠,手術(shù)暴露圓窗膜,用80umol/L哇巴因滴注于蒙古沙鼠耳蝸圓窗膜上,建立選擇性損傷螺旋神經(jīng)節(jié)細胞的動物模型。在成功建模的基礎(chǔ)上于蒙古沙鼠圓窗膜上滴注神經(jīng)營養(yǎng)因子-3(15ug/ml),穩(wěn)定7天后處死,立即取出耳蝸,制作病理切片進行HE染色,觀察Ⅰ型螺旋神經(jīng)節(jié)細胞數(shù)目、形態(tài)的改變,并用人工計數(shù)法采用SPSS17.0軟件進行統(tǒng)計學(xué)分析;制作電鏡切片,在透射電鏡下觀察細胞的超微結(jié)構(gòu),分析神經(jīng)營養(yǎng)因子-3對Ⅰ型螺旋神經(jīng)節(jié)細胞的保護作用。 結(jié)果: 本實驗用哇巴因成功建立了選擇性損傷螺旋神經(jīng)節(jié)細胞的動物模型,發(fā)現(xiàn)哇巴因?qū)Β裥吐菪窠?jīng)節(jié)細胞有明顯的損傷作用,損傷部位主要位于細胞膜周圍髓鞘組織、細胞核及線粒體,哇巴因組細胞計數(shù)較人工外淋巴液組明顯減少(P0.05);神經(jīng)營養(yǎng)因子-3對Ⅰ型螺旋神經(jīng)節(jié)細胞的細胞膜周圍髓鞘組織、線粒體及細胞核均有保護作用,在保護組細胞計數(shù)明顯高于對照組(P0.05)。 結(jié)論: 哇巴因損傷蒙古沙鼠Ⅰ型螺旋神經(jīng)節(jié)細胞后,經(jīng)圓窗膜滴注神經(jīng)營養(yǎng)因子-3(NT-3)對Ⅰ型螺旋神經(jīng)節(jié)細胞有一定的保護作用。對于臨床上尋找保護耳蝸聽神經(jīng)細胞的有效藥物及給藥途徑具有重要的價值。

The protective effect of neurotrophic factor-3 on cochlear spiral ganglion cells and its neuroprotective mechanism were studied.
