耳蝸局部應(yīng)用NR2B受體阻滯劑對水楊酸鈉致豚鼠螺旋神經(jīng)節(jié)細(xì)胞損傷的拮抗作用

【摘要】:目的通過圓窗龕局部給予特異性NR2B受體阻滯劑艾芬地爾,探討其拮抗水楊酸鈉致豚鼠螺旋神經(jīng)節(jié)細(xì)胞損傷的作用。方法將ABR閾值小于40dB SPL的健康花色豚鼠48只隨機(jī)分為4組,每組12只,Ⅰ組為空白對照組;Ⅱ組為人工外淋巴液(artificial perilymph,APL)組,左耳圓窗龕注人APL 60μl;Ⅲ組為水楊酸鈉組,左耳圓窗龕注入APL 60μl,腹腔注射水楊酸鈉;IV組為艾芬地爾組,左耳圓窗龕注入溶于APL的艾芬地爾(10μmol/L)60μl后,腹腔注射水楊酸鈉。水楊酸鈉注射量為400mg·kg-1·d-1,連續(xù)注射7天。停藥并行ABR測試后將動物處死,取出左側(cè)耳蝸,每組隨機(jī)取6只動物進(jìn)行免疫組織化學(xué)染色觀察耳蝸螺旋神經(jīng)節(jié)(SGN)caspase-3表達(dá)情況,6只行原位末端轉(zhuǎn)移酶標(biāo)記技術(shù)(terminal deoxynucleotidyl transferase-mediated dUTPbiotin nick end labeling assay,TUNEL)法檢測螺旋神經(jīng)節(jié)細(xì)胞凋亡率。結(jié)果給藥后,I、II、III、IV組動物左耳ABR反應(yīng)閾分別為31.67±5.16、33.33±5.17、64.17±7.36、49.17±5.85dB SPL;免疫組織化學(xué)染色顯示,Ⅰ、Ⅱ組螺旋神經(jīng)節(jié)細(xì)胞中caspase-3的表達(dá)量差異無統(tǒng)計學(xué)意義,Ⅲ、IV組螺旋神經(jīng)節(jié)細(xì)胞中caspase-3的表達(dá)量較Ⅰ、Ⅱ組顯著增高(P0.01),IV組螺旋神經(jīng)節(jié)細(xì)胞中caspase-3的表達(dá)量較Ⅲ組明顯降低(P0.01)。TUNEL法檢測結(jié)果示,Ⅰ、Ⅱ組SGN細(xì)胞凋亡率無顯著差異,Ⅲ、IV組SGN細(xì)胞凋亡率較Ⅰ、Ⅱ組顯著增高(P0.01),IV組SGN細(xì)胞凋亡率較Ⅲ組明顯降低(P0.01)。結(jié)論耳蝸局部特異性阻斷NR2B受體可以拮抗水楊酸鈉致豚鼠螺旋神經(jīng)節(jié)細(xì)胞的損傷。
