【摘要】：目的評(píng)估兔成視網(wǎng)膜細(xì)胞瘤(retinoblastoma,RB)模型的建立及經(jīng)導(dǎo)管眼動(dòng)脈灌注(selective ophthalmic arterial injection,SOAI)治療兔RB的可行性。方法 HXO-Rb44RB細(xì)胞予中心細(xì)胞室培養(yǎng),取對(duì)數(shù)生長(zhǎng)期HXO-Rb44細(xì)胞,用PBS配成濃度為2.0×107個(gè)/ml的細(xì)胞懸液,抽取0.1 ml細(xì)胞懸液注射于6只SPF級(jí)新西蘭兔右眼視網(wǎng)膜間隙下。接種腫瘤細(xì)胞前3 d和注射后1個(gè)月臀部注射環(huán)孢素A 15 mg·kg-1·d-1,1個(gè)月后減量至10 mg·kg-1·d-1并持續(xù)到實(shí)驗(yàn)結(jié)束。將RB兔分為對(duì)照組3只和實(shí)驗(yàn)組3只,對(duì)照組灌注0.9%氯化鈉溶液15 ml,實(shí)驗(yàn)組將美法侖2.5 mg用0.9%氯化鈉溶液稀釋至15 ml后灌注。術(shù)后分別于7、14 d比較兩組腫瘤生長(zhǎng)情況。結(jié)果注射環(huán)孢素A期間,實(shí)驗(yàn)兔不良反應(yīng)輕微,6只均建模成功。7次插管中6次成功插至眼動(dòng)脈。SOAI術(shù)后1周,實(shí)驗(yàn)組腫瘤可見明顯縮小,2周后均消失。SOAI術(shù)后,實(shí)驗(yàn)組僅1只見右眼輕度紅腫、流淚,1周后紅腫自行消退。結(jié)論 SOAI治療RB兔具有可行性,為進(jìn)一步經(jīng)眼動(dòng)脈灌注治療RB的基礎(chǔ)研究提供了參考。
[Abstract]:Objective to evaluate the establishment of rabbit retinoblastoma (retinoblastoma,RB) model and the feasibility of transcatheter ophthalmic artery infusion of (selective ophthalmic arterial injection,SOAI in the treatment of rabbit RB. Methods HXO-Rb44RB cells were cultured in the center cell chamber. The HXO-Rb44 cells in logarithmic growth phase were collected and injected into the right retinal space of 6 SPF grade New Zealand rabbits with PBS admixture of 2.0 脳 107 cells / ml. Cyclosporine A 15 mg kg-1 d -1 was injected into the buttocks 3 days before inoculation and 1 month after injection, and then decreased to 10 mg kg-1 d -1 one month later and continued until the end of the experiment. RB rabbits were divided into control group (n = 3) and experimental group (n = 3). The control group was perfused with 0.9% sodium chloride solution for 15 ml,. The tumor growth was compared between the two groups on the 7th and 14th day after operation. Results during the period of cyclosporine A injection, the adverse reaction of experimental rabbits was slight, and 6 rabbits were successfully modeled, 6 out of 7 intubation were successfully inserted into the ophthalmic artery. 1 week after SOAI, the tumor in the experimental group was obviously reduced and disappeared 2 weeks later. SOAI operation was performed. In the experimental group, the right eye was mild redness and tears, and the swelling subsided 1 week later. Conclusion SOAI is feasible for the treatment of RB rabbits, which provides a reference for the further study of transocular arterial infusion of RB.
