順鉑對鼻咽癌細胞rad51基因表達的影響

【摘要】:目的:通過檢測RAD51基因表達情況,比較人鼻咽癌高分化鱗癌細胞(CNE1)、低分化鱗癌細胞(CNE2)與順鉑治療的敏感性,探討DNA修復(fù)基因RAD51是否能成為順鉑治療鼻咽癌的有效靶點,并在其發(fā)生發(fā)展中起到的作用。 方法:(1)采用Real-time quantitative PCR (RT-PCR)技術(shù)檢測兩組不同分化程度的鼻咽癌細胞與正常鼻粘膜上皮細胞中RAD51基因的表達情況,以及使用順鉑處理后的表達情況;(2)采用MTT方法檢測并比較順鉑對不同分化程度鼻咽癌細胞的細胞存活率的影響。 結(jié)果:(1)鼻咽癌高分化鱗癌細胞(CNE1)和鼻咽癌低分化鱗癌細胞(CNE2)中RAD51的mRNA水平均明顯高于正常鼻粘膜組織細胞(P0.01),差異有統(tǒng)計學(xué)意義。(2)三個劑量組(4μmol/L組、8μmol/L組、16μmol/L組)順鉑干預(yù)鼻咽癌細胞后, CNE2細胞中RAD51的表達高于CNE1細胞,以4μmol/L組和8μmol/L組為甚(P0.05),差異有統(tǒng)計學(xué)意義。(3)同時,,各劑量組CNE1的細胞存活率,均低于CNE2(P0.01),差異有統(tǒng)計學(xué)意義。 結(jié)論:RAD51在分化程度不同的鼻咽癌細胞中表達不同,在化療藥物刺激下,分化程度越低的鼻咽癌細胞,RAD51的表達越高,對順鉑化療的敏感性越低。
[Abstract]:Objective: to compare the sensitivity of highly differentiated squamous cell carcinoma (CNE1), poorly differentiated squamous cell carcinoma (CNE2) and cisplatin in human nasopharyngeal carcinoma (NPC) by detecting the expression of RAD51 gene. To investigate whether DNA repair gene RAD51 can be an effective target for cisplatin in the treatment of nasopharyngeal carcinoma and its role in the pathogenesis and development of nasopharyngeal carcinoma. Methods: (1) Real-time quantitative PCR (RT-PCR technique was used to detect the expression of RAD51 gene in nasopharyngeal carcinoma (NPC) cells and normal nasal epithelial cells with different differentiation degree, and the expression of RAD51 gene after cisplatin treatment. (2) the effect of cisplatin on the survival rate of nasopharyngeal carcinoma cells with different differentiation was detected and compared by MTT. Results: (1) the mRNA levels of RAD51 in highly differentiated squamous cell carcinoma (CNE1) and poorly differentiated squamous cell carcinoma (CNE2) of nasopharyngeal carcinoma were significantly higher than those in normal nasal mucosa (P0.01). (2) after three dose groups (4 渭 mol/L group, 8 渭 mol/L group, 16 渭 mol/L group) treated with cisplatin, the expression of RAD51 in CNE2 cells was higher than that in CNE1 cells, especially in 4 渭 mol/L group and 8 渭 mol/L group (P0.05). (3) at the same time, the cell survival rate of CNE1 in each dose group was lower than that of CNE2 (P0.01), and the difference was statistically significant. Conclusion: the expression of RAD51 in nasopharyngeal carcinoma cells with different differentiation degree is different. Under the stimulation of chemotherapeutic drugs, the lower the differentiation degree, the higher the expression of RAD51 and the lower the sensitivity to cisplatin chemotherapy.
