喉鱗狀細(xì)胞癌中CyclinE和p27的表達(dá)和臨床意義

【摘要】:目的研究CyclinE和p27在喉鱗狀細(xì)胞癌(laryngeal squamous cell carcinoma,LSCC)中的表達(dá)及其相關(guān)性,并探討其與LSCC患者病情進(jìn)展之間的關(guān)系。 方法1、應(yīng)用逆轉(zhuǎn)錄-多聚酶鏈反應(yīng)(Reverse Transcription PCR, RT-PCR)技術(shù)對(duì)14例LSCC及癌旁正常喉黏膜組織(normal laryngeal tissues, NLT)中CyclinE和p27基因進(jìn)行擴(kuò)增,并檢測(cè)上述組織中二者的mRNA水平;2、免疫組織化學(xué)Elivision染色法檢測(cè)160例LSCC和對(duì)照組30例NLT中CyclinE和p27蛋白的表達(dá)水平,X2檢驗(yàn)分析CyclinE和p27在LSCC及NLT的表達(dá)差異及其與LSCC臨床病理因素之間的關(guān)系,Spearman等級(jí)相關(guān)分析CyclinE和p27之間的關(guān)系,P0.05為差異有統(tǒng)計(jì)學(xué)意義。 結(jié)果1、CyclinE和p27mRNA在LSCC和NLT中均有陽(yáng)性表達(dá)。LSCC和NLT組織中CyclinE mRNA的相對(duì)表達(dá)量分別為1.8007±0.1063和0.8458±0.1230,組間比較差異有統(tǒng)計(jì)學(xué)意義(P0.05);p27mRNA的相對(duì)表達(dá)量分別為0.6510±0.1451和1.5216±0.1923,組間比較差異有統(tǒng)計(jì)學(xué)意義(P0.05);2、CyclinE和p27蛋白在LSCC和NLT中表達(dá)水平不同。LSCC和NLT組織中,CyclinE蛋白陽(yáng)性率分別為64.37%(103/160)和16.67%(5/30); p27蛋白陽(yáng)性率分別為40%(64/160)和60%(18/30),組間比較差異有統(tǒng)計(jì)學(xué)意義(P0.05),,LSCC中CyclinE和p27的表達(dá)與腫瘤分化程度、淋巴結(jié)轉(zhuǎn)移和臨床分期有關(guān)(P0.05),與年齡、性別及直徑無(wú)關(guān)(P0.05);Spearman相關(guān)性分析顯示CyclinE和p27之間呈顯著負(fù)相關(guān)(R=-0.542,P0.05)。 結(jié)論: 1. CyclinE和p27mRNA在LSCC和NLT中均呈陽(yáng)性表達(dá),在LSCC中CyclinEmRNA顯著上調(diào),p27mRNA顯著下降。 2. CyclinE蛋白在LSCC中高表達(dá),在NLT中低表達(dá)甚至不表達(dá);p27蛋白在LSCC中低表達(dá)甚至不表達(dá),在NLT中呈高表達(dá)。 3. CyclinE和p27mRNA與CyclinE蛋白和p27蛋白在LSCC和NLT中表達(dá)一致。 4. CyclinE的表達(dá)水平與腫瘤分化程度、淋巴結(jié)轉(zhuǎn)移和臨床分期呈正相關(guān),LSCC進(jìn)展的重要指標(biāo)。p27的表達(dá)水平與腫瘤分化程度、淋巴結(jié)轉(zhuǎn)移和臨床分期呈負(fù)相關(guān),是LSCC進(jìn)展的重要指標(biāo)。
[Abstract]:Objective to investigate the expression of CyclinE and p27 in laryngeal squamous cell carcinoma (laryngeal squamous cell carcinoma,LSCC) and their correlation with the progression of LSCC. Methods 1. The CyclinE and p27 genes were amplified by reverse transcription-polymerase chain reaction (Reverse Transcription PCR, RT-PCR) in 14 cases of LSCC and adjacent normal laryngeal mucosa, and their mRNA levels were detected. 2. The expression of CyclinE and p27 protein in 160 cases of LSCC and 30 cases of control group were detected by immunohistochemical Elivision staining. The expression of CyclinE and p27 in LSCC and NLT and their relationship with the clinicopathological factors of LSCC were analyzed by X2 test. Spearman grade correlation analysis of the relationship between CyclinE and p27, P0.05 for the difference was statistically significant. Results 1CyclinE and p27mRNA were positive in LSCC and NLT, the relative expression of CyclinE mRNA in LSCC and NLT were 1.8007 鹵0.1063 and 0.8458 鹵0.1230respectively, the difference was statistically significant (P0.05). The relative expression of p27mRNA was 0.6510 鹵0.1451 and 1.5216 鹵0.1923, respectively, and the difference was statistically significant (P0.05). 2the expression levels of cyclin E and p27 protein in LSCC and NLT were different. The positive rates of CyclinE protein in LSCC and NLT were 64.37% (103 / 160) and 16.67% (5 / 30), respectively. The positive rates of p27 protein were 40% (64 / 160) and 60% (18 / 30), respectively. The difference between the two groups was statistically significant (P0.05). The expression of CyclinE and p27 in), LSCC was related to tumor differentiation, lymph node metastasis and clinical stage (P0.05). It was not related to age, sex and diameter (P0.05). Spearman correlation analysis showed that there was a significant negative correlation between CyclinE and p27 (P 0.05). Conclusion: 1. Both CyclinE and p27mRNA were positive in LSCC and NLT, CyclinEmRNA was up-regulated in LSCC, and p27mRNA was significantly decreased. 2. CyclinE protein was overexpressed in LSCC, low or non-expressed in NLT, and p27 protein was overexpressed in LSCC and NLT. 3. The expression of CyclinE and p27mRNA was consistent with that of CyclinE and p27 in LSCC and NLT. 4. The expression of CyclinE was positively correlated with the degree of tumor differentiation, lymph node metastasis and clinical stage. The expression level of p27 was negatively correlated with the degree of tumor differentiation, lymph node metastasis and clinical stage, and was an important indicator of the progression of LSCC.
