【摘要】： 目的：通過檢測抑癌基因Annexin 1在喉鱗狀細(xì)胞癌(LSCC)及癌旁不同距離組織中的表達(dá)情況,從蛋白水平及基因水平來探討其與喉癌手術(shù)安全切緣的關(guān)系,并分析與臨床分型、臨床分期、病理分級和淋巴結(jié)轉(zhuǎn)移的關(guān)系。 方法：采用病理學(xué)觀察、免疫組織化學(xué)法和RT-PCR法檢測32例新鮮喉鱗癌標(biāo)本的癌原發(fā)灶區(qū)與癌旁黏膜交界處0.2cm、0.5cm、1.0cm、2.0cm組織中Annexin 1的表達(dá)情況。 結(jié)果：Annexin 1 (ANXAl)在距離喉鱗癌原發(fā)灶0.2cm→2.0cm中的表達(dá)強度依次遞增；在癌旁0.5cm以外組織中,Annexin 1蛋白的染色強度很深,而在喉鱗癌原發(fā)灶及癌旁0.2cm組織中,Annexin 1蛋白的染色強度相對較低,兩者之間統(tǒng)計學(xué)差異有顯著性(P0.01)。 免疫組化技術(shù)分析Annexin 1的表達(dá)與臨床分期(P0.05)、病理分級(P0.05)均有關(guān)；與臨床分型、淋巴結(jié)轉(zhuǎn)移均無顯著性差異(P0.05); RT-PCR法測定Annexin lmRNA的表達(dá)與喉鱗癌的臨床分期(P0.05)、病理分級(P0.05)有關(guān)；與臨床分型、淋巴結(jié)轉(zhuǎn)移均無關(guān)(P0.05)。 結(jié)論：Annexin 1蛋白及1mRNA在LSCC組織中表達(dá)量由原發(fā)灶到無癌區(qū)的遞增性變化及癌旁0.5cm內(nèi)外表達(dá)量的顯著性差異表達(dá),提示：Annexin 1可以作為LSCC惡性程度的分子生物學(xué)指標(biāo),指導(dǎo)臨床病理分級及分期；癌旁0.5cm做為手術(shù)切緣是安全的。
[Abstract]:Objective: to investigate the expression of tumor suppressor gene Annexin 1 in laryngeal squamous cell carcinoma (LSCC) (LSCC) and its adjacent tissues at different distances from the level of protein and gene, and to analyze the relationship between the expression of Annexin 1 gene and the safe margin of laryngeal carcinoma operation, and to analyze the relationship between the expression and clinical type of laryngeal squamous cell carcinoma (LSCC). The relationship between clinical stage, pathological grade and lymph node metastasis. Methods: the expression of Annexin 1 in 32 cases of fresh laryngeal squamous cell carcinoma (LSCC) was detected by pathological observation, immunohistochemical method and RT-PCR method. Results: the expression intensity of Annexin 1 (ANXAl) in 0.2cm 2.0cm was increased in turn, and the staining intensity of Annexin 1 protein was very strong in the adjacent 0.5cm tissues of laryngeal squamous cell carcinoma (LSCC). In laryngeal squamous cell carcinoma (LSCC) and adjacent 0.2cm tissues, the staining intensity of Annexin 1 protein was relatively low, there was significant difference between them (P0.01). The expression of Annexin 1 was correlated with clinical stage (P0.05) and pathological grade (P0.05), and had no significant difference with clinical type and lymph node metastasis (P0.05). The expression of Annexin lmRNA was correlated with the clinical stage (P0.05) and pathological grade (P0.05) of laryngeal squamous cell carcinoma (P0.05). There was no correlation with clinical type and lymph node metastasis (P0.05). Conclusion: the expression of Annexin 1 protein and 1mRNA in LSCC tissues is increased from primary tumor to non-cancerous area, and there is significant difference in the expression of 0.5cm in and out of LSCC, suggesting that Annexin 1 can be used as a molecular biological index of the malignant degree of LSCC. It is safe to use the paracancerous 0.5cm as the surgical margin to guide the clinicopathological grading and staging.
