苦參堿對(duì)人鼻咽癌CNE2細(xì)胞裸鼠移植瘤生長及凋亡相關(guān)基因表達(dá)的影響

【摘要】:目的:觀察苦參堿對(duì)人鼻咽癌CNE2細(xì)胞裸鼠移植瘤生長及凋亡相關(guān)基因表達(dá)的影響。方法:20只裸鼠皮下接種CNE2,建立移植瘤模型,成瘤后,隨機(jī)分成4組,每組5只?鄥A高、低劑量組分別腹腔注射100、50mg/kg苦參堿,陽性對(duì)照組腹腔注射順鉑2 mg/kg,陰性對(duì)照組給予生理鹽水。給藥7次后,應(yīng)用體內(nèi)抑瘤實(shí)驗(yàn)測定抑瘤率,分別采用real-time PCR法和Western blot法檢測移植瘤組織中bax、bcl-2、caspase-3 mRNA和蛋白的表達(dá)。結(jié)果:陽性對(duì)照組和苦參堿低、高劑量組抑瘤率分別為51.6%和26.4%、43.2%。4組間移植瘤組織中bax、bcl-2、caspase-3的mRNA和蛋白表達(dá)差異有統(tǒng)計(jì)學(xué)意義(F=3.751、16.113、3.918和26.500、9.771、12.255,P0.05);與陰性對(duì)照組比較,陽性對(duì)照組和苦參堿高、低劑量組bax mRNA和蛋白表達(dá)及陽性對(duì)照組、苦參堿高劑量組caspase-3 mRNA和蛋白表達(dá)上調(diào)(P0.05),bcl-2 mRNA和蛋白表達(dá)下調(diào)(P0.05)。結(jié)論:苦參堿可通過調(diào)節(jié)CNE2移植瘤組織中凋亡相關(guān)基因的表達(dá)抑制腫瘤的生長。
[Abstract]:Aim: to observe the effect of matrine on the growth and apoptosis-related gene expression of human nasopharyngeal carcinoma (NPC) CNE2 cells in nude mice. Methods: 20 nude mice were inoculated subcutaneously with CNE2, to establish transplanted tumor model. After tumorigenesis, 20 nude mice were randomly divided into 4 groups, 5 in each group. Matrine was injected intraperitoneally into 100g / kg matrine in low dose group, and normal saline was injected intraperitoneally in positive control group with cisplatin 2 mg/kg, negative control group. After seven times of administration, the tumor inhibition rate was measured by tumor inhibition test in vivo, and the expression of bax,bcl-2,caspase-3 mRNA and protein in transplanted tumor tissues were detected by real-time PCR and Western blot methods respectively. Results: the inhibitory rates of bax,bcl-2,caspase-3 in the positive control group and matrine group were 51.6% and 26.4%, respectively. There were significant differences in the expression of mRNA and protein in the transplanted tumor tissues between the two groups (F3. 751, 16.113, 3.918 and 26.500, 9.771, 12.255, P0.05), and compared with the negative control group, the positive control group and matrine group were higher than those in the negative control group, the positive control group and the matrine group were higher than those in the negative control group, the positive control group and the matrine group were significantly higher than those in the negative control group. The expression of bax mRNA and protein in low dose group and positive control group, the expression of caspase-3 mRNA and protein in high dose group of matrine was up-regulated (P0.05), and the expression of bcl-2 mRNA and protein was down-regulated (P0.05). Conclusion: matrine can inhibit the growth of CNE2 by regulating the expression of apoptosis-related genes.
