【摘要】:目的:分析L-多巴脫羧酶(DDC)mRNA和蛋白在喉癌組織中的表達(dá),以評(píng)估其在喉癌診斷和預(yù)后中的臨床意義。方法:從53例喉癌患者的腫瘤切除手術(shù)中獲取106份組織樣品,并從這些組織樣品中分別分離出總RNA。采用RT-PCR法對(duì)DDC mRNA的表達(dá)水平進(jìn)行量化,同時(shí)采用Western Blot蛋白質(zhì)印跡法對(duì)DDC蛋白的表達(dá)水平進(jìn)行檢測。結(jié)果:與正常喉黏膜組織相比,原發(fā)性喉癌(P=0.000)和復(fù)發(fā)性喉癌(P=0.033)樣品中的DDC mRNA表達(dá)水平都出現(xiàn)了顯著下調(diào)。且DDC mRNA表達(dá)水平與患者年齡和組織學(xué)分級(jí)無關(guān),但與TNM分期之間存在顯著的負(fù)相關(guān)性(P=0.021)。同時(shí),我們在腫瘤和正常喉組織中都檢測到了DDC蛋白的表達(dá)。結(jié)論:DDC的表達(dá)狀態(tài)可能在喉癌的發(fā)展中發(fā)揮重要的作用,在未來的喉癌患者臨床管理中具有重要的意義。
[Abstract]:Objective: to analyze the expression of L- dopa decarboxylase (DDC) mRNA and protein in laryngeal carcinoma and to evaluate its clinical significance in the diagnosis and prognosis of laryngeal carcinoma. Methods: 106 tissue samples were obtained from 53 patients with laryngeal carcinoma. The expression level of DDC mRNA was quantified by RT-PCR and the expression level of DDC protein was detected by Western Blot Western blot. Results: compared with normal laryngeal mucosa, the expression of DDC mRNA in primary laryngeal carcinoma (P0. 000) and recurrent laryngeal carcinoma (P0. 033) was significantly down-regulated. There was no correlation between DDC mRNA expression and age and histological grade, but there was a negative correlation between DDC mRNA expression and TNM stage (P0. 021). At the same time, we detected the expression of DDC protein in both tumor and normal laryngeal tissues. Conclusion the expression of DDC may play an important role in the development of laryngeal carcinoma and play an important role in the clinical management of laryngeal cancer patients in the future.
