【摘要】：目的：研究清瘤亮喉方對喉咽癌移植瘤生物學行為的影響,并探討其治療或干預喉咽癌的作用機制。方法：1.建立梨狀窩兔VX2瘤模型兔：VX2瘤細胞株荷瘤新西蘭大白兔傳代,以傳代兔腫瘤組織塊,制成腫瘤組織液,直達喉鏡下接種于兔梨狀窩。2.研究清瘤亮喉方對喉咽癌移植瘤生物學行為的影響：新西蘭大白兔46只接種腫瘤后,隨機分為空白組、對照組(氟尿嘧啶組)、治療組(清瘤亮喉方組),除7只種植兔意外死亡外,余39只進入研究。隨后觀察其生存情況、影像學(CT)、解剖學、病理學(HE染色)及免疫組化(細胞周期調(diào)控因子cyclinB、cyclinD、CDK4及P27)研究。結(jié)果：服用清瘤亮喉方組(治療組)及氟尿嘧啶組(對照組)的梨狀窩VX2瘤兔的生存質(zhì)量及存活時間明顯優(yōu)于空白組(p0.05)；治療組與對照組的梨狀窩VX2瘤兔的腫瘤生長情況在解剖及影像學上與空白組存在顯著差異(p0.05)；治療組與對照組的梨狀窩VX2瘤兔的腫瘤生長情況在病理學上與空白組存在顯著差異(p0.05)；治療組與對照組、空白組的梨狀窩VX2瘤兔的腫瘤生長情況在免疫組化研究上存在顯著差異(p0.05)。結(jié)論：1.清瘤亮喉方能有效改善梨狀窩VX2瘤兔的生存質(zhì)量,延長腫瘤兔的生命。2.清瘤亮喉方能干預喉咽癌移植瘤的生長,其機制可能是通過誘導腫瘤細胞凋亡。3.清瘤亮喉方對喉咽癌的治療作用,體現(xiàn)了中藥對機體免疫功能的調(diào)節(jié)。
[Abstract]:Objective: to study the effect of Qinglianghou recipe on the biological behavior of transplanted laryngopharyngeal carcinoma, and to explore the mechanism of its treatment or intervention in laryngopharyngeal carcinoma. Method 1: 1. The rabbit VX2 tumor model of piriform nest rabbit was established. The rabbit VX2 tumor cell line was subcultured into rabbit tumor tissue fluid and inoculated directly into rabbit piriform fossa under laryngoscope. To study the effect of Qinglianghou recipe on the biological behavior of transplanted laryngopharyngeal carcinoma, 46 New Zealand white rabbits were randomly divided into blank group after inoculation of tumor. The control group (fluorouracil group), the treatment group (Qingliulianghou prescription group), except 7 implanted rabbits died accidentally, the other 39 entered the study. Then, the survival status, imaging CTU, anatomy, pathology and HE staining were observed, and immunohistochemistry (cyclinBon cyclin DCDK4 and P27) were studied. Results: the quality of life and survival time of piriform fossa VX2 tumor rabbits in the treatment group and fluorouracil group were significantly better than those in the blank group (P 0.05), and the tumorigenesis of the piriform fossa VX2 tumor rabbits in the treatment group and the control group was significantly better than that in the control group. There were significant differences in anatomy and imaging between the treatment group and the blank group (p 0.05A), the growth of the VX2 tumor in the piriform fossa between the treatment group and the control group was significantly different from that in the blank group (P 0.05), and the tumor growth in the treatment group and the control group was significantly different from that in the blank group. There was a significant difference in tumor growth of piriform fossa VX2 tumor rabbits in the blank group in immunohistochemical study. Conclusion 1. Qingliangyao prescription can effectively improve the quality of life of VX2 tumor rabbits in piriform fossa and prolong the life of tumor rabbits. Qinglianghou prescription can interfere with the growth of transplanted tumor of laryngopharyngeal carcinoma, and its mechanism may be by inducing apoptosis of tumor cells. 3. The therapeutic effect of Qinglianghou prescription on laryngopharynx carcinoma reflects the regulation of immune function of traditional Chinese medicine.
