CK19和EGFR mRNA在喉癌動物模型外周血中的表達及其與淋巴轉(zhuǎn)移關(guān)系

【摘要】:目的:探索外周血中的循環(huán)播散腫瘤細胞(circulation tumor cell,CTC)與喉癌淋巴結(jié)轉(zhuǎn)移的相關(guān)性,評價CK19和EGFR在喉癌微轉(zhuǎn)移診斷中的價值。 方法:從40只裸鼠中隨機選擇35只為實驗組,平均分為5組,每組7只,建立喉癌裸鼠成瘤模型,其余5只為空白對照組。于接種后2、4、6、8、10周分別處死1組,采用RT-PCR方法檢測裸鼠外周血及腫瘤組織中CK19 mRNA和EGFR mRNA表達,采用連續(xù)病理切片HE染色法觀察淋巴結(jié)轉(zhuǎn)移情況,免疫組化SP法檢測腫瘤組織中CK19和EGFR蛋白的表達。 結(jié)果:裸鼠成瘤模型構(gòu)建成功。喉癌裸鼠動物模型中有20只發(fā)生淋巴結(jié)轉(zhuǎn)移,轉(zhuǎn)移率為57.1%(20/35)?瞻讓φ战M外周血中未發(fā)現(xiàn)CK19 mRNA和EGFR mRNA表達,實驗組外周血和腫瘤組織中CK19 mRNA陽性率分別為54.3%(19/35)和80%(28/35),EGFR mRNA的陽性率分別為37.1%(13/35)和74.3%(26/35),CK19和EGFR是CTC的特異性標記物,淋巴結(jié)轉(zhuǎn)移與外周血CTC相關(guān)(P0.001),且多發(fā)生在腫瘤指數(shù)生長期。免疫組化檢測結(jié)果發(fā)現(xiàn)CK19和EGFR蛋白在腫瘤組織中陽性率分別為80%(28/35)和85.7%(30/35)。 結(jié)論:外周血中CK19和EGFR的表達有助于預(yù)測淋巴結(jié)轉(zhuǎn)移,而CK19和EGFR是外周血循環(huán)播散腫瘤細胞的特異性標記物,提示CTC與喉癌淋巴轉(zhuǎn)移具有相關(guān)性。
[Abstract]:Objective: to investigate the relationship between circulating tumor cell CTCs and lymph node metastasis in laryngeal carcinoma, and to evaluate the value of CK19 and EGFR in the diagnosis of micrometastasis of laryngeal carcinoma.Methods: 35 nude mice were randomly divided into 5 groups, 7 in each group, to establish tumorigenic model of laryngeal carcinoma, and the other 5 were blank control group.The expression of CK19 mRNA and EGFR mRNA in peripheral blood and tumor tissues of nude mice were detected by RT-PCR method, and lymph node metastasis was observed by HE staining in serial sections.Immunohistochemical SP method was used to detect the expression of CK19 and EGFR in tumor tissues.Results: the tumorigenic model of nude mice was successfully constructed.Lymph node metastasis occurred in 20 nude mice with laryngeal carcinoma, and the metastasis rate was 57.1%.No expression of CK19 mRNA and EGFR mRNA was found in the peripheral blood of the blank control group. The positive rates of CK19 mRNA in peripheral blood and tumor tissues in the experimental group were 54.3 / 35 and 80 / 28 / 35, respectively. The positive rates of CK19 mRNA in peripheral blood of the control group were 37.1T / 35) and 74.33.33% / 35% / CK19 and EGFR were specific markers of CTC, respectively.Lymph node metastasis was associated with peripheral blood CTC (P 0.001), and most of them occurred in the growth stage of tumor index.Immunohistochemical results showed that the positive rates of CK19 and EGFR in tumor tissues were 80 / 28 / 35) and 85.730 / 35 / 35 respectively.Conclusion: the expression of CK19 and EGFR in peripheral blood is helpful to predict lymph node metastasis, while CK19 and EGFR are specific markers for circulating tumor cells in peripheral blood, suggesting that CTC is associated with lymphatic metastasis of laryngeal carcinoma.
