【摘要】：目的:研究雷公藤紅素對(duì)大鼠變應(yīng)性鼻炎(AR)的治療作用并初步探討相關(guān)作用機(jī)制。方法:構(gòu)建大鼠AR模型,測(cè)定大鼠第1天、第4天和第7天的行為學(xué)特征改變、鼻黏膜呼吸區(qū)組織超氧化物歧化酶(SOD)、丙二醛(MDA)、還原型谷胱甘肽(GSH)和谷胱甘肽過(guò)氧化物酶(GSH-PX)的水平;測(cè)定鼻黏膜呼吸區(qū)組織核因子E2p45相關(guān)因子2(NRF2)的核內(nèi)蛋白表達(dá)及谷氨酸半胱氨酸連接酶催化亞基(GCLC)的漿蛋白表達(dá)。結(jié)果:AR大鼠第1、4、7天的行為學(xué)特征改變顯著(P0.01),鼻黏膜呼吸區(qū)組織SOD、GSH、GSH-PX降低,MDA升高;NRF2的核內(nèi)蛋白表達(dá)降低,GCLC的漿蛋白表達(dá)降低。雷公藤紅素低劑量治療組和高劑量治療組能不同程度改善上述指標(biāo)的變化,同時(shí)提高了鼻黏膜呼吸區(qū)組織NRF2的核蛋白表達(dá)和GCLC的漿蛋白表達(dá)。結(jié)論:雷公藤紅素能改善AR大鼠的癥狀,其機(jī)制可能與提高鼻黏膜呼吸區(qū)組織NRF2的核蛋白表達(dá)和GCLC的漿蛋白表達(dá)相關(guān)。
[Abstract]:Objective: To study the effect of tripterine on rats with allergic rhinitis (AR) treatment and explore the related mechanism. Methods: the AR rat model, rats were first days, fourth days and seventh days of behavior change, nasal respiratory tissue superoxide dismutase (SOD). Malondialdehyde (MDA), glutathione (GSH) and glutathione peroxidase (GSH-PX) levels; Determination of nasal respiratory tissue nuclear factor E2p45 related factor 2 (NRF2) of the nuclear protein expression and glutamate cysteine ligase catalytic subunit (GCLC) expression of plasma protein. Results: the behavior of day 1,4,7 AR the characteristics of rats changed significantly (P0.01), nasal respiratory tissue SOD, GSH, GSH-PX decreased, MDA increased; the expression of nuclear protein NRF2 decreased expression of GCLC protein decreased. The plasma tripterine low dose group and high dose treatment group can be improved on Changes in the index, while improving the nuclear protein expression and GCLC nasal respiratory tissue NRF2 plasma protein expression. Conclusion: celastrol can improve the symptoms of AR rats, and its mechanism may improve nuclear protein expression and GCLC nasal respiratory tissue NRF2 plasma protein expression.
