氟他胺誘導(dǎo)干眼模型大鼠角膜和淚腺組織病理變化及TNF-α、IL-1α的表達(dá)

【摘要】:目的通過雄激素受體拮抗劑氟他胺溶液灌胃建立大鼠干眼模型,觀察干眼大鼠角膜及淚腺組織病理變化及TNF-α、IL~(-1)α的表達(dá)。方法取32只SPF級SD雌性大鼠隨機(jī)分為4組,氟他胺高、中、低濃度組和對照組,每組8只,每天分別用濃度為16 mg·kg-1、13 mg·kg-1、10 mg·kg-1氟他胺溶液和1 m L生理鹽水灌胃。給藥后0周、2周、4周、8周、16周、20周分別檢測各組基礎(chǔ)淚液分泌試驗(yàn)(SchirmerⅠtest,SⅠt)值、淚膜破裂時間(break-up time,BUT)及角膜熒光素鈉染色情況。給藥后0周、10周、20周采用放射免疫法檢測各組血清睪酮濃度。給藥后20周處死大鼠取各組角膜與淚腺組織行病理學(xué)觀察,免疫組織化學(xué)方法檢測各組TNF-α、IL~(-1)α的表達(dá)情況。結(jié)果給藥后4周時高、中濃度組SⅠt值開始減小,與對照組比較差異均有統(tǒng)計學(xué)意義(均為P0.05)。8周時高、中、低濃度組的SⅠt值均小于對照組(均為P0.05),20周時高(3.23±0.31)mm、中(3.98±0.36)mm、低(4.73±0.18)mm濃度組的SⅠt值進(jìn)一步減小,且均較對照組(9.60±0.60)mm下降大于50%(均為P0.05),高、中、低濃度組兩兩比較差異均有統(tǒng)計學(xué)意義(均為P0.05)。給藥后4周時高濃度組大鼠即出現(xiàn)BUT縮短,8周時,高、中、低濃度組的BUT均短于對照組(均為P0.05),20周時高(2.40±0.61)s、中(4.32±0.68)s、低(6.10±0.81)s濃度組的BUT較對照組(9.12±0.73)s進(jìn)一步縮短,各組之間兩兩比較差異均有統(tǒng)計學(xué)意義(均為P0.05)。給藥后4周時高、中濃度組開始出現(xiàn)角膜上皮點(diǎn)狀熒光素鈉著色,16周時中、低濃度組出現(xiàn)小片狀著色,高濃度組可見角膜上皮彌漫性大片狀著色,20周時各組染色評分兩兩比較差異均有統(tǒng)計學(xué)意義(均為P0.05)。給藥后20周時睪酮濃度高濃度組(5.08±0.21)pg·m L~(-1)、中濃度組(4.23±0.16)pg·m L~(-1)、低濃度組(3.58±0.24)pg·m L~(-1)較對照組(2.24±0.15)pg·m L~(-1)不同程度升高,與對照組比較差異均有統(tǒng)計學(xué)意義(均為P0.05),高、中、低濃度組兩兩比較差異均有統(tǒng)計學(xué)意義(均為P0.05)。HE染色顯示高、中、低濃度組角膜出現(xiàn)不同程度鱗狀上皮層次增加,基底細(xì)胞排列紊亂,基質(zhì)層水腫;高、中、低濃度組淚腺上皮細(xì)胞小而密集,腺泡萎縮融合,結(jié)構(gòu)紊亂,高濃度組改變最為顯著。免疫組織化學(xué)結(jié)果顯示高、中、低濃度組角膜與淚腺上皮細(xì)胞中TNF-α、IL~(-1)α陽性表達(dá)細(xì)胞數(shù)均高于對照組(均為P0.05)。結(jié)論雄激素受體拮抗劑氟他胺溶液灌胃可以成功誘導(dǎo)混合型干眼模型,干眼大鼠角膜和淚腺上皮細(xì)胞發(fā)生病理學(xué)改變,炎性因子TNF-α、IL~(-1)α的表達(dá)明顯增高。
