AhR和TGF-β1在伴或不伴過敏性鼻炎的鼻息肉中表達(dá)差異初步研究

【摘要】:研究背景:很多種因素參與慢性鼻-鼻竇炎(chronic rhino sinusitis,CRS)的組織重構(gòu)過程,這其中就包括過敏因素、轉(zhuǎn)化生長因子β1(Transforming Growth Factor-beta 1,TGF-β1)等。另外,芳香烴受體(aryl hydrocarbon receptor,AhR)也參與其中,但是目前其具體機(jī)制還尚不明確。目的:本研究的目的就是要通過檢測AhR與TGF-β1在CRS中的表達(dá)來探究兩者與過敏狀態(tài)和組織重構(gòu)的可能相關(guān)性。方法:收集四組病人共75例,包括CRS不伴鼻息肉(nasal polyps,NP)不伴過敏性鼻炎(allergic rhinitis,AR)組(CRSs NPs AR)20例,CRS伴鼻息肉伴AR組(CRSwNPw AR)20例,CRSwNP不伴AR組(CRSwNPs AR)20例和對照組15例。采用馬松三色染色方法,來評價40例患者中膠原的表達(dá)情況;通過免疫組織化學(xué)染色(SP法)來觀察AhR和TGF-β1在CRS中的分布與表達(dá)水平;及通過實時熒光定量檢測方法(q RT-PCR)和蛋白質(zhì)免疫印跡法(Western blotting)分別檢測AhR和TGF-β1的核酸和蛋白質(zhì)表達(dá)水平。結(jié)果:通過組織學(xué)染色,以及在核酸水平和蛋白質(zhì)水平進(jìn)行檢測分析發(fā)現(xiàn),在CRSs NPs AR組,膠原水平、AhR和TGF-β1的表達(dá)均高于CRSwNPs AR(p0.05)。但是,CRSwNPw AR與CRSwNPs AR相比,膠原水平和TGF-β1是增高的,而AhR表達(dá)水平是降低的。AhR和TGF-β1的表達(dá)與膠原表達(dá)水平呈現(xiàn)正相關(guān)性。結(jié)論:我們推測AhR也許與TGF-β1一樣也參與到了CRSwNP的組織重構(gòu)中。而在CRSwNPw AR中AhR的低表達(dá)可能與過敏性反應(yīng)有關(guān)。AhR與TGF-β1在CRS組織重構(gòu)中的相互作用還有待進(jìn)一步研究探討。
[Abstract]:Background: there are many factors involved in the process of tissue remodeling in chronic rhinosinusitis (chronic rhino sinusitis,CRS), including allergic factors, transforming growth factor 尾 1 (Transforming Growth Factor-beta 1 (TGF- 尾 1) and so on. In addition, the aromatic receptor (aryl hydrocarbon receptor,AhR is also involved, but its mechanism is still unclear. Aim: to explore the possible correlation between AhR and TGF- 尾 1 in CRS by detecting the expression of TGF- 尾 1 in CRS. Methods: 75 patients in four groups were collected, including 20 patients with CRS without nasal polyps (nasal polyps,NP) without allergic rhinitis (allergic rhinitis,AR), 20 patients with CRS with nasal polyps with AR (CRSwNPw AR), 20 patients with CRSwNP without AR (CRSwNPs AR) and 15 patients with control group (control group). The expression of collagen in 40 patients was evaluated by using Ma Song three-color staining method, and the distribution and expression of AhR and TGF- 尾 1 in CRS were observed by immunohistochemical staining (SP). The expression levels of nucleic acid and protein in AhR and TGF- 尾 1 were detected by real-time fluorescence quantitative detection (Q RT-PCR) and Western blot (Western blotting), respectively. Results: the expression of AhR and TGF- 尾 1 in CRSs NPs AR group was higher than that in CRSwNPs AR (group (p0.05). However, compared with CRSwNPs AR, the collagen level and TGF- 尾 1 in, CRSwNPw AR were increased, while the AhR expression was decreased. There was a positive correlation between the expression of AhR and TGF- 尾 1 and collagen expression. Conclusion: we speculate that AhR may be involved in the tissue remodeling of CRSwNP as well as TGF- 尾 1. However, the low expression of AhR in CRSwNPw AR may be related to allergic reaction. The interaction between AhR and TGF- 尾 1 in CRS tissue remodeling needs further study.
